摘要
目的探讨黄芩苷基于白介素(interleukin,IL)-23/IL-17轴对支气管扩张大鼠肺功能的影响和作用机制。方法选取65只SD大鼠,10只作为对照组不作任何干预,其余通过注射铜绿假单胞菌构建支气管扩张大鼠模型,5只大鼠用于确认造模结果,余50只大鼠将随机分为模型组、黄芩苷低剂量组(50 mg/kg)、中剂量组(100 mg/kg)、高剂量组(200 mg/kg)和阳性对照组(左氧氟沙星,96.7 mg/kg),每组10只。干预14 d后,检测各组大鼠肺功能指标;酶联免疫吸附(enzyme linked immunosorbent assay,ELISA)法检测血清中IL-23、IL-17水平以及肺组织中基质金属蛋白酶(matrix metalloproteinase,MMP)9、组织金属蛋白酶抑制物(tissue inhibitor of metalloproteinase,TIMP)-1含量;苏木精-伊红(hematoxylin-eosin,HE)染色观察各组大鼠肺组织病理学变化;免疫印迹法(western blot,WB)检测各组大鼠肺组织中IL-23、IL-17蛋白表达水平。结果与模型组相比,黄芩苷低、中、高剂量组大鼠肺组织炎性细胞浸润、支气管扩张、肺泡结构受损等均得到不同程度缓解,肺功能指标和TIMP-1含量升高(P<0.01),MMP-9、IL-23、IL-17水平及IL-23、IL-17蛋白表达均降低(P<0.01)。黄芩苷高剂量组各项指标与阳性对照组差异无统计学意义(P>0.05)。结论黄芩苷可能通过抑制IL-23/IL-17轴,降低支气管扩张大鼠炎症水平,减少肺组织损伤,提高肺功能。
Objective To investigate the effects and mechanism of baicalin on pulmonary function in bronchiectasis rats based on interleukin(IL)-23/IL-17 axis.Methods A total of 65 SD rats were selected,10 of which were used as the control group without any intervention,and the rest rats were injected with Pseudomonas aeruginosa to construct the bronchiectasis rat model.Five rats were used to confirm the modeling results.The remaining 50 rats were randomly divided into model group,low-dose(50 mg/kg),medium-dose(100 mg/kg),high-dose(200 mg/kg)baicalin group and positive control(levofloxacin,96.7 mg/kg)group,with 10 rats in each group.After 14 days of intervention,pulmonary function indexes of rats in each group were measured.The levels of IL-23 and IL-17 in serum and the contents of matrix metalloproteinase(MMP)9 and tissue inhibitor of metalloproteinase(TIMP)-1 in lung tissue were detected by enzyme linked immunosorbent assay(ELISA).Hematoxylin and eosin(HE)staining was used to observe the pathological changes of lung tissue in each group.Western blot(WB)was used to detect the protein expression levels of IL-23 and IL-17 in the lung tissue of rats in each group.Results Compared with the model group,the inflammatory cell infiltration,bronchiectasis,alveolar structure damage in the lung tissue of rats in the low-dose,medium-dose and high-dose baicalin groups were relieved to varying degrees,the lung function index and TIMP-1 content were increased(P<0.01),and the levels of MMP-9,IL-23,IL-17 and the expression of IL-23,IL-17 protein were decreased(P<0.01).There were no significant differences in the indexes between the high-dose baicalin group and the positive control group(P>0.05).Conclusion Baicalin may reduce the inflammatory level and lung tissue injury in bronchiectasis rats and improve lung function by inhibiting the IL-23/IL-17 axis.
