LINC00511调控NF-κB通路对多囊卵巢综合征卵巢颗粒细胞凋亡和炎性反应的影响 被引量:2

摘要 目的探讨LINC00511在多囊卵巢综合征中的表达及其对卵巢颗粒细胞(KGN)凋亡、炎性反应的影响和机制。方法RT-qPCR检测LINC00511在KGN和人正常卵巢上皮细胞IOSE-80中的表达。将KGN细胞分为si-NC组、si-LINC00511组、pcDNA-LINC00511组、pcDNA组和si-LINC00511+TNF-α组。流式细胞术评估KGN细胞凋亡率。ELISA法检测细胞培养液中白介素-6(IL-6)和IL-1β水平。蛋白质印迹法检测磷酸化核因子κB(NF-κB)、p65(pp65)蛋白表达。结果KGN细胞中LINC00511表达水平显著高于IOSE-80细胞(P<0.05)。与si-NC组比较,si-LINC00511组KGN细胞凋亡率显著升高(P<0.05),p-p65蛋白表达、细胞培养液中IL-6和IL-1β水平显著降低(P<0.05)。与pcDNA组比较,pcDNA-LINC00511组KGN细胞凋亡率显著降低(P<0.05),p-p65蛋白表达、细胞培养液中IL-6和IL-1β水平显著升高(P<0.05)。与si-LINC00511组比较,si-LINC00511+TNF-α组KGN细胞凋亡率显著降低(P<0.05),p-p65蛋白表达、细胞培养液中IL-6和IL-1β水平显著升高(P<0.05)。结论干扰LINC00511通过抑制NF-κB可促进多囊卵巢综合征卵巢颗粒细胞凋亡,抑制细胞炎性反应。 Objective To investigate the expression and effects of LINC00511 on the apoptosis and inflammatory reaction of ovarian granulosa cells(KGN),and to explore its action mechanism.Methods The expression levels of LINC00511 in KGN cells and human normal ovarian epithelial cells-IOSE-80 were detected by RT-qPCR.And KGN cells were divided into si-NC group,si-LINC00511 group,pcDNA-LINC00511 group,pcDNA group and si-LINC00511+TNF-αgroup.The apoptosis rate of KGN cells was detected by flow cytometry.The levels of IL-6 and IL-1βin cell culture medium were detected by ELISA,and Western Blot was performed to detect the expression levels of phosphorylated nuclear factor kappa B(NF-κB)p65(p-p65)protein.Results The expression levels of LINC00511 in KGN cells were significantly higher than those in IOSE-80 cells(P<0.05).Compared with that in si-NC group,the apoptosis rate of KGN cells in si-LINC00511 group was significantly increased(P<0.05),however,the expression levels of p-p65 protein,IL-6 and IL-1βin cell culture medium were significantly decreased(P<0.05).Compared with that in pcDNA group,the apoptosis rate of KGN cells in pcDNA-LINC00511 group was significantly decreased(P<0.05),but,and the expression levels of p-p65 protein,IL-6 and IL-1βin cell culture medium were significantly increased(P<0.05).Compared with that in si-LINC00511 group,the apoptosis rate of KGN cells in si-LINC00511+TNF-αgroup was significantly decreased(P<0.05),however,the expression levels of p-p65 protein,IL-6 and IL-1βin cell culture medium were significantly increased(P<0.05).Conclusion Interfering with LINC00511 can the apoptosis of KGN and inhibit the cellular inflammatory reaction by inhibiting the expression of NF-κB pathway.
