OPN TIMP-2及STAT3蛋白在子宫内膜异位症患者中的表达及临床意义 被引量:3

摘要 目的:分析骨桥蛋白(osteopontin,OPN)、基质金属蛋白酶组织抑制因子2(Tissue inhibitor of matrix metalloproteinase 2,TIMP-2)及信号转导和转录激活因子3(Signal transducer and activator of transcription 3,STAT3)蛋白在子宫内膜异位症(Endometriosis,EMS)患者中的表达及临床意义。方法:选取2019年1月至2020年1月在本院收治的117例子宫内膜异位症患者临床资料[EMS组,根据美国生育协会评分标准(r-AFS)分期,Ⅰ~Ⅱ期53例,Ⅲ~Ⅳ期64例;月经周期:增生期66例,分泌期51例],另选取同期收治子宫肌瘤或不孕症行手术治疗的非EMS者103例(对照组)。对比不同人群、不同r-AFS分期以及不同月经周期中OPN、TIMP-2及STAT3蛋白表达情况,分析OPN、TIMP-2及STAT3与r-AFS分期相关性。结果:EMS组OPN、STAT3表达明显高于对照组,TIMP-2水平则明显低于对照组(P<0.05);Ⅲ~Ⅳ期OPN、STAT3表达明显高于Ⅰ~Ⅱ期,TIMP-2水平显低于Ⅰ~Ⅱ期(P<0.05);增生期OPN、STAT3表达明显高于分泌期,TIMP-2水平则明显低于分泌期(P<0.05);EMS患者血清OPN、STAT3与r-AFS分期为正相关,TIMP-2与r-AFS分期为负相关(P<0.05)。结论:在EMS患者中OPN、STAT3表达增加、TIMP-2表达下调,三者与患者发生与病情进展间关系密切,监测三者表达情况可为疾病发展预测及转归提供参考。 Objective:To analyze the expression and clincal significance of osteopontin(OPN),tissue inhibitor of matrix metalloproteinase 2(TIMP-2)and signal transducer and activator of transcription 3(STAT3)proteins in endometriosis(Endometriosis,EMS)patients.Methods:Clinical data of 117 patients with endometriosis admitted to our hospital from January 2019 to January 2020 were selected[EMS group,according to the American Fertility Society scoring standard(r-AFS)staging,53 cases in stagesⅠ-Ⅱand 64 cases in stagesⅢ-Ⅳ;menstrual cycle:66 cases in the proliferative phase and 51 cases in the secretory phase],and another 117 cases admitted to the same period with uterine 103 non-EMS patients with fibroids or infertility treated surgically(control group)were selected.The expression of OPN,TIMP-2 and STAT3 in different populations,different r-AFS stages and different menstrual cycles were compared,and the correlation between OPN,TIMP-2 and STAT3 and r-AFS stages was analyzed.Results:The expression of OPN and STAT3 in EMS group was significantly higher than that in control group,while the level of TIMP-2 was significantly lower than that in control group(P<0.05);the expression of OPN and STAT3 in stageⅢ~Ⅳwas significantly higher than that in stageⅠ-Ⅱ,while the level of TIMP-2 was significantly lower than that in stageⅠ-Ⅱ(P<0.05);the expression of OPN and STAT3 in proliferative stage was significantly higher than that in secretory stage,while the TIMP-2 levels were significantly lower than those in the secretory phase(P<0.05);serum OPN and STAT3 were positively correlated with r-AFS stage,and TIMP-2 was negatively correlated with r-AFS stage in EMS patients(P<0.05).Conclusion:OPN and STAT3 expression were increased and TIMP-2 expression was down-regulated in EMS patients,which were closely related to the occurrence and progression of the disease,and monitoring their expression could provide a reference for disease development prediction and regression.
