摘要
为了解湖南省猪圆环病毒3型(PCV3)的遗传变异情况,以25份PCV3阳性样品DNA为模板,使用PCR方法分两段扩增PCV3全基因序列并进行拼接。结果获得7株PCV3分离株全基因组,3株PCV3 ORF2序列。7株PCV3全基因组的核苷酸序列同源性为98.8%~99.7%,10株ORF2的核苷酸序列同源性为97.8%~99.8%。根据系统发育树,PCV3可分为3个分支,所有分离株的遗传关系均较近。ORF2氨基酸序列突变少,发现2个在其他参考毒株中未出现的突变位点。研究获得的PCV3毒株序列与国内外PCV3毒株序列其遗传关系较近,无特殊突变,较为稳定。
In order to understand the genetic variation of porcine circovirus type 3(PCV3) in Hunan province, using 25 PCV3 positive samples DNA as template, the PCV3 genome sequences were amplified by PCR.Results showed that the genomes of 7 PCV3 isolates and the PCV3 ORF2 sequences of 3 isolates were obtained, the genomes of 7 PCV3 isolates were all 2 000 bp in size.The nucleotide sequence homologies of the genomes of 7 PCV3 strains ranged from 98.8% to 99.7%,and the nucleotide sequence homologies of 10 strains of ORF2 ranged from 97.8% to 99.8%.According to the phylogenetic tree, PCV3 can be divided into three subtypes, and the genetic relationships of all isolates are close.The ORF2 amino acid sequence has few mutations.Two mutation sites that did not appear in other reference strains were found.The genetic relationship of PCV3 obtained in this study is close to that of PCV3 at home and abroad, and there is no special mutation, so it is relatively stable.
