摘要
为探讨柑橘黄龙病碘—淀粉快速检测法(IR)的专化性及其影响因素,本研究采集大量柑橘黄龙病疑似症状样品,以及柑橘衰退病、裂皮病、碎叶病、天牛危害、柑橘生理缺锌、缺钙6种常见柑橘病害感病植株样品,进行碘—淀粉快速检测、淀粉含量测定、柑橘黄龙病PCR检测。发现碘—淀粉快速检测的33个样品中仅2个植株样品检错,IR检测与常规PCR检出结果的一致率较高,且发现柑橘天牛危害与柑橘裂皮病是影响IR检测专化性的主要因素。因此,柑橘黄龙病田间碘—淀粉快速检测专化性较高,结合田间症状诊断及生物、环境等影响因素综合分析,可进一步提高其专化性。
In order to explore the specificity and influencing factors of the rapid detection method of iodine starch (IR) for Citrus Huanglongbing, a large number of suspected symptomatic samples of Citrus Huanglongbing, as well as six common citrus disease susceptible plant samples of citrus recessive disease, peel disease, leaf crush disease, longicorn beetle damage, physiological zinc deficiency and calcium deficiency of citrus were collected for rapid detection of iodine starch, determination of starch content and PCR detection of Citrus Huanglongbing. It was found that only 2 plant samples in 33 samples detected by iodine starch rapid detection were wrong, and the consistency between IR detection and conventional PCR detection was high. It was found that citrus longicorn harm and citrus peel disease were the main factors affecting the specificity of IR detection. Therefore, the specificity of rapid detection of Citrus Huanglongbing in the field is high. Combined with the diagnosis of field symptoms and the comprehensive analysis of biological, environmental and other influencing factors, the specificity can be further improved.
