摘要
目的:采用超高效液相色谱-四级杆-飞行时间串联质谱法(UPLC-Q-TOF/MS/MS)鉴定长柄臭黄荆醇提物化学成分。方法:采用Waters ACQUITY UPLC HSS T3 (2.1 mm×100 mm, 1.8μm)色谱柱,流动相(0.1%甲酸水溶液-乙腈)梯度洗脱,使用AB Sciex Triple TOF■ 4600高分辨质谱。基于ESI正、负离子采集模式获得MS并进行数据分析。在此基础上,根据成分保留时间结合Natural Products HR-MS/MS Spectral Library 1.0数据库进行匹配及相关文献报道进行结构鉴定。结果:从长柄臭黄荆醇提物中共鉴定出53个化合物,其中黄酮类19种,酚酸类16种,苯乙醇类5种,脂肪酸类6种,杂环类化合物7种。结论:UPLC-Q-TOF/MS/MS可快速鉴定长柄臭黄荆醇提物的化学成分,为长柄臭黄荆质量控制、发现新化合物和有价值的药用成分提供依据。
Objective To identify the chemical constituents of the ethanol extract of Premna puberula Pamp. using ultra-performance liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry(UPLC-Q-TOF/MS/MS). Methods Waters ACQUITY UPLC HSST3 column(2.1 mm×100 mm, 1.8 μm) was used with the gradient elution using mobile phase(0.1% formic acid aqueous solution-acetonitrile) and AB Sciex Triple TOF4600 high resolution mass spectrometry(MS). Based on ESI positive and negative ions acquisition mode, the MS data was obtained and analyzed. On this basis, the structure was identified according to the retention time of components and the database of Natural Products HR-MS/MS Spectral Library 1.0 and relevant literature reports. Results A total of 53 compounds were identified from the alcohol extract of Premna puberula Pamp., including 19 flavonoids, 16 phenolic acids, 5 Phenylethanol, 6 fatty acids and 7 heterocyclic compounds. Conclusion UPLC-Q-TOF/MS/MS can rapidly identify the chemical constituents of the alcoholic extract of Premna Puberula Pamp., and provide a basis for quality control, discovery of new compounds and valuable medicinal components of Premna puberula Pamp.
