旱辣蓼质量标准研究

摘要 建立旱辣蓼的质量标准。方法:通过显微鉴别和TLC法定性鉴别旱辣蓼;按《中国药典》检查水分、总灰分、酸不溶性灰分、水溶性浸出物、醇溶性浸出物、重金属及有害元素;HPLC法测定没食子酸的含量。结果:考查确定了旱辣蓼的显微特征;通过TLC法能清晰显示芦丁对照品特征斑点;10批旱辣蓼中水分含量平均值为8.8%;总灰分含量平均值为11.7%;酸不溶性灰分含量平均值为2.5%;水溶性浸出物含量平均值为21.0%;醇溶性浸出物含量平均值为7.6%;铅、镉、砷、汞、铜的含量平均值分别为1.8、0.22、0.5、0.00和36.0 mg/kg;采用高效液相色谱法检测没食子酸含量,没食子酸在3.845~76.9μg/ml范围内呈良好的线性关系(r=0.999 6),平均加样回收率为97.2%,RSD为2.62%。结论:该方法准确可信,操作简便可行,为旱辣蓼药材的质量控制提供了理论依据。 Objective:In this study,the quality standard control method of Polygonum flaccidum Meisn.was established.Methods:Microscopic identification and TLC were adopted in the qualitative identification of P.flaccidum.The contents of water,total ash,acid-insoluble ash,water-soluble extractive,alcohol-soluble extract,heavy metals,and harmful elements were determined according to the methods of Chinese Pharmacopoeia.The content of gallic acid was determined by HPLC.Results:The microscopic features of P.flaccidum were identified.The characteristic spots of the rutin reference substance could be clearly displayed by TLC.The average contents of water,total ash,insoluble acid ash,water-soluble extract,and alcohol-soluble extract in ten batches of P.flaccidum were 8.8%,11.7%,2.5%,21.0%,and 7.6%,respectively.The average contents of Pb,Cd,As,Hg,and Cu were 1.8,0.22,0.5,0.00 mg/kg,and 36.0 mg/kg,respectively.The content of gallic acid was determined by high-performance liquid chromatography.Gallic acid showed a good linear relationship in the 3.845 ~76.9 μg/ml(r=0.999 6),with an average recovery of 97.2% and RSD of 2.62%.Conclusion:This method is accurate and reliable,and the operation is simple and feasible,which provides a theoretical basis for the quality control of P.flaccidum.
