甘蔗细胞壁蔗糖转化酶基因SoCIN2的克隆和表达分析 被引量:1

Cloning and Expression Analysis of Cell Wall Invertase Gene SoCIN2 in Sugarcane

摘要 细胞壁蔗糖转化酶(CIN)主要参与韧皮部质外体的蔗糖卸载,在调控果实发育中起作用。为探究CIN在调控甘蔗糖分积累过程中的作用,本试验从低糖甘蔗品种B8中克隆到一个1个CIN基因,命名为SoCIN2基因。结果显示,SoCIN2基因全长为2269 bp,其中开放阅读框为1857 bp,编码618个氨基酸。SoCIN2蛋白分子量为67.59 kD,理论等电点为5.25;定位于细胞壁上,不含信号肽序列;具有保守的NDPNG、FRDP和MWECP结构域,属于糖基转移酶GH32家族。系统进化分析表明,甘蔗SoCIN2蛋白与高粱SbCIN2遗传距离最近。荧光定量PCR结果表明,在甘蔗根、茎、叶和芽中SoCIN2基因均表达,其中在叶中基因表达量最高,茎中最低。甘蔗+1叶中SoCIN2基因表达量显著高于茎中。在伸长期和成熟期,茎中SoCIN2基因的表达模式相同,成熟茎中基因表达量显著高于未成熟茎中基因表达量,表明SoCIN2基因可能在调控甘蔗成熟茎中蔗糖积累中起作用。 Cell wall invertase(CIN)is mainly involved in the unloading of sucrose from phloem apoplasts and plays a role in regulating fruit development.In order to explore the function of CIN in the regulation of sucrose accumulation in sugarcane,a CIN gene was cloned from the low-sugar sugarcane variety B8 and named as SoCIN2 gene.The results showed that the full length of SoCIN2 gene was 2269 bp,of which the open reading frame was 1857 bp,encoding 618 amino acids.The molecular weight of SoCIN2 protein is 67.59 kD,and the theoretical isoelectric point is 5.25.It is located on the cell wall and does not contain a signal peptide sequence.It has conserved NDPNG,FRDP and MW ECP domains and belongs to the GH32 family of glycosyltransferases.Phylogenetic analysis showed that SoCIN2 and SbCIN2 have the closest genetic distance.Fluorescence quantitative PCR results showed that the expression of SoCIN2 gene could be detected in roots,stems,leaves and shoots of sugarcane,with the highest expression in leaves and the lowest in stems.In different growth stages of sugarcane,the expression level of SoCIN2 gene in+1 leaves was significantly higher than that in immature stems.The expression pattern of SoCIN2 gene in stems was the same in elongation and mature stages of sugarcane,and the expression level of SoCIN2 gene in mature stems was significantly higher than that in immature stems,indicating that SoCIN2 gene may play a role in regulating sucrose accumulation in mature stems of sugarcane.
