摘要
本研究建立了高效液相色谱定量检测薰衣草花中花色苷的方法。薰衣草花样品经酸化乙醇超声提取,采用C18色谱柱,流动相为1%甲酸水溶液(A)和1%甲酸乙腈溶液(B),检测波长为530 nm,花色苷含量以飞燕草素-3-O-葡萄糖苷的含量表示。结果表明:飞燕草素-3-O-葡萄糖苷在1~100μg/m L浓度范围内,浓度与峰面积线性关良好(R^(2)=0.9999)。对3个不同时期的两个薰衣草品种进行花色素含量检测,发现颜色越深的薰衣草飞燕草素-3-O-葡萄糖苷的含量越多,其含量色多少与颜色呈正相关,且随着薰衣草的花发育成熟,飞燕草素-3-O-葡萄糖苷的含量也逐渐增多。本研究结果为进一步探讨薰衣草花色形成的生化途径和开展薰衣草花色的改良育种等研究提供了参考。
A method for the quantitative determination of anthocyanins in lavender flowers by high performance liquid chromatography(HPLC)was established.The lavender flower samples were ultrasonically extracted with acidified ethanol,using a C18 chromatographic column,the mobile phase was 1% formic acid aqueous solution(A)and 1%formic acid acetonitrile solution(B),the detection wavelength was 530 nm.The content of anthocyanins was expressed by the content of delphinin-3-o-glucoside.The results showed that the linear range of del phinidin-3-o-glucoside was 1~100μg/mL(R^(2)=0.9999).The anthocyanin content of two Lavender varieties in three different periods was detected.It was found that the deeper the color,the higher the content of delphinin-3-o-glucoside and the deeper the color.The content of delphinin-3-o-glucoside gradually increased with the development and maturity of lavender flowers.The results of this study provide a reference for further research on the biochemical pathways of lavender flower color formation and the improvement and breeding of lavender flower color.
