不同粒径牛乳脂肪球中磷脂酰胆碱含量的质谱分析

Mass spectrometry analysis of phosphatidylcholine content in cow’s milk fat globules with different sizes

摘要 为了探究不同粒径乳脂肪球层与磷脂酰胆碱(PC)和溶血磷脂酰胆碱(LPC)含量的关系,试验采用重力分层法对中国荷斯坦奶牛乳样进行分层,用激光粒度仪测量不同层乳样的粒径参数{表面积相关等效直径(D_([3,2]))、体积相关的等效直径(D_([4,3]))、10%粒子的体积径[Dv(10)]、50%粒子的体积径[Dv(50)]、90%粒子的体积径[Dv(90)]、比表面积(SSA),用液相色谱串联质谱分析乳脂肪球粒径最大层和最小层乳样中PC和LPC含量。结果表明:乳样在重力因素下从下到上共分为1~7层(F1~F7);粒径参数D_([3,2])、D_([4,3])、Dv(10)、Dv(50)、Dv(90)均呈现从F1到F7逐渐增大的趋势,而SSA呈现逐渐下降的趋势;F1的D_([3,2])和D_([4,3])最小,分别为2.11μm和2.37μm,显著小于其他层(P<0.05);F7 D_([3,2])和D_([4,3])最大,分别为3.37μm和4.16μm,显著大于其他层(P<0.05)。在F1(粒径最小层)和F7(粒径最大层)乳样中共检测到18种PC和17种LPC;与F1乳样相比,F7层乳样中PC和LPC含量均增加,其中PC(28∶0)、PC(30∶0)、PC(31∶0)、PC(32∶0)、PC(34∶0)、PC(32∶1)、PC(34∶1)、PC(36∶1)、PC(32∶2)、PC(34∶2)、PC(36∶2)、PC(34∶3)、PC(36∶3)、PC(36∶4)、PC(38∶5)、PC(38∶6)、LPC(14∶0/0∶0)、LPC(15∶0/0∶0)、LPC(16∶0/0∶0)、LPC(17∶0/0∶0)、LPC(16∶1/0∶0)、LPC(17∶1/0∶0)、LPC(18∶1/0∶0)、LPC(18∶2/0∶0)、LPC(20∶2/0∶0)、LPC(18∶3/0∶0)、LPC(20∶3/0∶0)、LPC(20∶4/0∶0)、LPC(22∶4/0∶0)、LPC(22∶5/0∶0)含量均显著升高(P<0.05)。说明牛乳在重力分层条件下小粒径脂肪球层的PC和LPC含量低于大粒径脂肪球层。 In order to explore the relationship between milk fat globules with different particle sizes and phosphatidylcholine(PC)and lyso-phosphatidylcholine(LPC)content,the milk samples of Chinese Holstein cows were stratified by gravity stratification method.The particle size parameters of different layers of milk samples{surface area related equivalent diameter(D_([3,2])),volume related equivalent diameter(D_([4,3])),volume size of 10%particles[Dv(10)],volume size of 50%particles[Dv(50)],volume size of 90%particles[Dv(90)]and specific surface area(SSA)were measured by laser granulometer.The contents of PC and LPC in milk fat globules with the largest and smallest particle sizes were analyzed by liquid chromatography tandem mass spectrometry.The results showed that the milk samples were divided into 1-7 layers(F1-F7)under gravity.From F1 layer to F7 layer,the particle size parameters such as D_([3,2]),D_([4,3]),Dv(10),Dv(50)and Dv(90)all showed a gradual increasing trend,while SSA showed a gradual decreasing trend.The values of D_([3,2])and D_([4,3])in F1 layer were the smallest,which were 2.11μm and 2.37μm,respectively,and significantly lower than other layers(P<0.05).The values of D_([3,2])and D_([4,3])in F7 floor were the largest.The values of D_([3,2])and D_([4,3])were 3.37μm and 4.16μm,respectively,and were significantly larger than other layers(P<0.05).A total of 18 kinds of PC and 17 kinds of LPC were detected in the milk samples of F1 layer and F7 layer.Compared with the contents of PC and LPC in the milk samples of F1 layer,the contents of PC and LPC in the milk samples of F7 layer were all increased,and the contents of PC(28∶0),PC(30∶0),PC(31∶0),PC(32∶0),PC(34∶0),PC(32∶1),PC(34∶1),PC(36∶1),PC(32∶2),PC(34∶2),PC(36∶2),PC(34∶3),PC(36∶3),PC(36∶4),PC(38∶5),PC(38∶6)),LPC(14∶0/0∶0),LPC(15∶0/0∶0),LPC(16∶0/0∶0),LPC(17∶0/0∶0),LPC(16∶1/0∶0),LPC(17∶1/0∶0),LPC(18∶1/0∶0),LPC(18∶2/0∶0),LPC(20∶2/0∶0),LPC(18∶3/0∶0),LPC(20∶3/0∶0),LPC(20∶4/0∶0),LPC(22∶4/0∶0),LPC(22∶5/0∶0)were significantly increased in the F7 layer(P<0.05).The results indicated that the PC and LPC contents in small-diameter fat globules were lower than those in large-diameter fat globules under gravity stratification.
