摘要
为研究蛋清中功能蛋白随鸡胚正常发育的动态变化,使用TMT标记定量技术检测0、4、8、12胚龄种蛋蛋清中的蛋白质,并进行蛋白质组学分析,筛选出在不同时间点差异表达的81个蛋白质。对这81个差异蛋白进行GO、Pathway和HCA分析,结果表明:在鸡胚早期发育过程中,蛋清中的大部分功能蛋白主要在脂质代谢、氧化还原反应、调节胚胎组织或器官发育这3个方面发挥相关作用。HCA分析结果表明蛋清中调控脂质代谢的蛋白质如卵黄蛋白原(VTG1、VTG2、VTG3)、载脂蛋白(APOB、APOA1、APOV1)等,主要在4~8胚龄开始发挥作用;调控氧化还原过程的血红蛋白(HBA1、HBZ、HBBR、HBAD、HBBA)主要在8胚龄之后发挥作用;调控胚胎组织器官发育的蛋白质则随着胚胎发育的不同阶段,在不同时间点起到相应作用。蛋清重要功能蛋白在鸡胚早期发育过程中呈动态变化,且具有各自独特的变化规律,结果对丰富胚胎发育的理论基础具有重要意义。
To study the dynamic changes of functional proteins in egg white, the proteins in the egg white of 0, 4, 8 and 12 embryonic days were detected by TMT labeling quantitative detection technique, and proteomic analysis was carried out. 81 proteins differentially expressed at different time points were screened out. GO, Pathway and HCA analysis of these 81 differential proteins showed that most of the functional proteins in egg white mainly played related roles in lipid metabolism, REDOX reaction, and regulation of embryonic tissue or organ development during early embryonic development. HCA analysis of 81 differential proteins showed that proteins regulating lipid metabolisms, such as vitellogenin(VTG1, VTG2, VTG3) and apolipoprotein(APOB, APOA1, APOV1), mainly played roles in ED4-ED8. The hemoglobin(HBA1, HBZ, HBBR, HBAD, HBBA) regulating the REDOX process mainly played a role after ED8. Proteins that regulated embryonic development acted at different time points in different stages of embryonic development. In conclusion, important functional proteins of egg white showed dynamic changes and had their own unique change during the early development of chicken embryo. This study was of great significance to enrich the theoretical basis of embryo development.
