摘要
本试验旨在探究槐定碱对白色念珠菌生物被膜形成的影响。通过微量稀释法测定槐定碱对白色念珠菌的最小抑菌浓度(MIC)及对该菌株生物被膜生长过程中不同时期的抑制率,使用光学显微镜和扫描电子显微镜观察槐定碱对白色念珠菌生物被膜形成的影响,通过牛乳平板法、卵黄平板法、SDS法检测槐定碱对白色念珠菌的胞外蛋白酶活性、磷脂酶活性及胞外DNA的影响。结果表明:槐定碱对白色念珠菌的MIC值为8 mg/mL,在光学显微镜下发现槐定碱对白色念珠菌生物被膜的形成有影响,尤其对生物被膜黏附期和生长期有明显的抑制作用,而对成熟期后抑制作用减弱。此外,槐定碱对白色念珠菌胞外DNA和胞外蛋白酶无影响,对其磷脂酶活性有显著抑制作用。
The purpose of this experiment was conducted to investigate effects of sophoridine on Candida albicans biofilm forma-tion.The minimum inhibitory concentration(MIC)and the inhibition rate of sophoridine on C.albicans biofilm in different periods were determined by microdilution method.The inhibition of sophoridine on C.albicans biofilm was observed by light microscope and scanning electron microscope.The effects of sophoridine on the extracellular protease activity,phospholipase activity and extra-cellular DNA of C.albicans were detected by milk plate method,egg-yolk plate method and SDS method,respectively.The results showed that the MIC was 8 mg/mL and sophoridine had an effect on the formation of C.albicans biofilm by light microscope,and had a significant inhibitory effect on the adhesion stage and growth stage of the biofilm,but the inhibitory effect weakened after the maturity stage.In addition,sophoridine had no effect on the extracellular DNA and extracellular protease of C.albicans,but inhibit-ed the phospholipase activity.
