摘要
目的建立高效液相色谱(HPLC)波长切换法同时测定补中益气丸(浓缩丸)中甘草苷、阿魏酸、异阿魏酸、橙皮苷、甘草酸铵的含量。方法样品采用70%甲醇溶液超声波提取,色谱柱为Ultimate XB-C18(4.6 mm×250 mm,5μm),柱温为30℃,流动相为乙腈-0.1%磷酸溶液,梯度洗脱,流速为1.0 ml/min,检测波长为276 nm(甘草苷)、324 nm(阿魏酸、异阿魏酸)、283 nm(橙皮苷)、252 nm(甘草酸铵)。结果甘草苷、阿魏酸、异阿魏酸、橙皮苷、甘草酸铵分别在7.858~392.886μg/ml(r=0.9996)、0.895~44.730μg/ml(r=1.0000)、6.880~343.975μg/ml(r=0.9999)、6.733~336.674μg/ml(r=0.9995)、30.630~1531.52μg/ml(r=0.9998)范围内与峰面积呈良好线性关系,回归方程分别为Y=0.2829X+0.1681,Y=0.3424X+0.0301,Y=0.3309X+0.2628,Y=0.1061X+0.3332,Y=0.0506X+1.1699,各成分平均加样回收率为97.06%~100.73%,RSD为1.13%~2.02%。结论该方法操作简单,重复性好,可为补中益气丸(浓缩丸)多指标质量评价及控制提供科学依据。
Objective To establish an HPLC multi-wavelength switching method for the simultaneous determination of liquiritin,ferulic acid,isoferulic acid,hesperidin and ammonium glycyrrhizinate in Buzhong Yiqi pills(concentrated).Methods Samples were extracted by ultrasonic method using 70%methanol solution.The analysis was performed on Ultimate XB-C18(4.6 mm×250 mm,5μm)column with acetonitrile-0.1%phosphoric acid solution as the mobile phase at 30℃at 1.0 ml/min in a gradient elution manner.The detection wavelength was set at 276 nm for liquiritin,324 nm for ferulic acid and isoferulic acid,283 nm for hesperidin,252 nm for ammonium glycyrrhizinate.Results Liquiritin,ferulic acid,isoferulic acid,hesperidin and ammonium glycyrrhizinate showed good linear relationships within the ranges of 7.858-392.886μg/ml(r=0.9996),0.895-44.730μg/ml(r=1.0000),6.880-343.975μg/ml(r=0.9999),6.733-336.674μg/ml(r=0.9995),30.630-1531.52μg/ml(r=0.9998),and the regression equations were Y=0.2829X+0.1681,Y=0.3424X+0.0301,Y=0.3309X+0.2628,Y=0.1061X+0.3332,Y=0.0506X+1.1699.The average recoveries were 97.06%-100.73%with the RSD of 1.13%-2.02%.Conclusion The established method is simple and reproducible,which can provide a scientific basis for the multi-index quality evaluation and the control of Buzhong Yiqi pills(concentrated).
