基于网络药理学和实验验证探讨薏苡附子败酱散治疗溃疡性结肠炎作用机制 被引量:7

Study on Mechanism of Yiyi Fuzi Baijiang Powder in Treatment of Ulcerative Colitis Based on Network Pharmacology and Experimental Verification

摘要 目的采用网络药理学方法预测薏苡附子败酱散治疗溃疡性结肠炎(UC)的作用机制,并通过动物实验对相关靶点进行初步验证。方法利用TCMSP、TCMIP、SwissADME、SwissTargetPrediction、UniProt数据库筛选薏苡附子败酱散活性成分及相关靶点,利用GeneCards、OMIM、DrugBank数据库获得UC相关靶点,获取二者共同靶点,使用Cytoscape软件构建药物-成分-靶点-疾病网络。将共同靶点导入STRING数据库得到蛋白相互作用网络,筛选核心靶点。使用OmicShare(基迪奥)在线平台进行GO功能富集分析,利用R软件进行KEGG通路富集分析。建立UC小鼠模型并用薏苡附子败酱散进行干预,对核心靶点及相关通路进行初步验证。结果筛选得到薏苡附子败酱散治疗UC的32个活性成分及100个靶点。GO富集分析显示,主要条目为细胞进程、代谢过程、生物调节、刺激反应等生物过程,连接、催化活性、分子功能调控等分子功能,细胞、细胞组分、细胞器等细胞组分。KEGG通路富集分析显示,涉及肝炎、病毒感染、癌症、AGE-RAGE、细胞凋亡、TNF、p53、PI3K/Akt等信号通路。实验结果表明,薏苡附子败酱散对UC小鼠的结肠病理有改善作用,Caspase3(半胱氨酸天冬氨酸蛋白酶3)及EGFR(表皮生长因子受体)是其作用靶点,PI3K/Akt是其作用通路之一。结论薏苡附子败酱散通过多靶点、多通路治疗UC,可为相关研究提供依据。 Objective To explore the mechanism of Yiyi Fuzi Baijiang Powder in the treatment of ulcerative colitis(UC)based on network pharmacology;To conduct preliminary verification of relevant targets through animal experiments.Methods Active components and related targets of Yiyi Fuzi Baijiang Decoction were screened out based on TCMSP,TCMIP,SwissADME,SwissTargetPrediction and Uniprot database.UC-related targets were obtained based on GeneCards,OMIM,DrugBank database,then the common targets in between were screened out.Cytoscape software was applied to construct ingredient-target-disease network.The common targets were entered into the STRING database to obtain the protein-protein interaction network,and the core targets were obtained.GO enrichment analysis was performed on OmicShare online platform,and KEGG pathway enrichment was analyzed by R software.A mice model of UC was established and intervened with Yiyi Fuzi Baijiang Powder to preliminarily verify the core targets and related pathways.Results Totally 32 active components and 100 UC-related targets were identified from Yiyi Fuzi Baijiang Powder.GO enrichment analysis showed that the main items were biological processes such as cellular processes,metabolic processes,biological regulation,and responses to stimulus,molecular functions such as binding,catalytic activity,and molecular function regulator,and cellular components such as cells,cell parts,and organelles.KEGG pathway enrichment analysis showed that hepatitis,virus infection,cancer,AGERAGE,apoptosis,TNF,P53,PI3K/Akt pathways were enriched.The experiment further proved that Yiyi Fuzi Baijiang Powder could improve the colonic pathology of UC mice.In addition,Caspase3 and EGFR were proved to be the therapeutic targets,and PI3K/Akt pathway was one of action pathways.Conclusion Yiyi Fuzi Baijiang Powder can provide a basis for related research in the treatment of UC through multiple targets and multiple pathways.
