1株降解牦牛血液菌株的分离鉴定 被引量:1

摘要 研究旨在解决屠宰场血液污染和血液利用问题,以甘肃省兰州市一屠宰场被血液浸润的土样为分离基质,分离鉴定能够有效降解牦牛血液的菌株。通过血平板筛选能够分解血液蛋白的菌株,对筛选的菌株进行形态学鉴定、生化反应试验、16S rRNA基因序列、蛋白酶活力测定及血液降解能力测定,筛选出1株能够高效降解血红蛋白的菌株。结果显示,该分离株为革兰阳性杆菌,命名为NWMCC0050。16S rRNA序列系统进化分析显示,菌株NWMCC0050与Aneurinibacillus migulanus strain NBRC 15520的序列相似性为99.85%,为解硫胺素芽孢杆菌。当牦牛血液添加量为30%时,该菌株对血液蛋白降解率达9.86%。研究表明,该菌株对牦牛血液具有一定的降解能力,具有作为利用废弃血液制备氨基酸肥料的候选功能菌株的潜力。 The purpose of the study was to solve the problems of blood pollution and blood utilization in slaughterhouses.The soil samples infiltrated by blood in a slaughterhouse in Lanzhou City,Gansu Province were used as the separation matrix to isolate and identify strains that can effectively degrade yak blood.The strains that can decompose blood proteins are screened by blood plate,and the screened strains are subjected to morphological identification,biochemical reaction test,16S r RNA gene sequence,protease activity determination and blood degradation ability determination.A strain that can efficiently degrade hemoglobin was screened.The results showed that the isolated strain was a gram-positive bacillus,named NWMCC0050.The phylogenetic analysis of the 16S rRNA sequence showed that the sequence similarity between the strain NWMCC0050 and Aneurinibacillus migulanus strain NBRC 15520 was 99.85%,and it was Bacillus thiamine solubilizer.When the amount of yak blood added was 30%,the degradation rate of blood protein by the strain reached9.86%.The study indicates that the strain has a certain ability to degrade yak blood and has the potential to be a candidate functional strain for preparing amino acid fertilizers from waste blood.
