摘要
目的:探讨国家专利数据库收载的中药复方专利治疗心力衰竭的配伍用药规律及核心药物的作用机制。方法:检索国家知识产权局专利检索及分析系统,收集2002年1月-2021年2月治疗心力衰竭的中药复方专利并建立数据库,对中药使用频次、药性、药味、归经、功效分类、主治证型进行统计,对常用药物进行Aprirori算法相关性分析和药物关联网络分析,探寻治疗疾病的核心药物。通过TCMSP数据库获得高频核心药物有效成分及作用靶点,应用CTD、OMIM和GeneCardes数据库检索疾病相关靶点。应用STRING数据库构建蛋白互作网络,应用DAVID数据库进行GO分析和KEGG信号通路分析,运用Cytoscape软件构建成分-靶点-信号通路网络。结果:收集到治疗心力衰竭中药复方专利161项,涉及中药432味,频次≥5的常用中药78味,频次≥21的高频中药18味,得到10个高频药对和10个高频角药。药物功效类别以补气药、活血药、利水渗湿药及温里药为主,核心药物为黄芪、丹参、茯苓、附子,网络药理学分析发现其治疗心力衰竭主要通过作用于IL-6、TNF、VEGFA、PTGS2、NOS2等潜在靶点,调控肿瘤坏死因子(TNF)信号通路、PI3K-AKT信号通路、缺氧诱导因子(HIF-1)信号通路等发挥作用。结论:中药复方专利治疗心力衰竭以补气活血、温阳利水的配伍为主,核心药物“黄芪、丹参、茯苓、附子”的作用机制可能是通过干预IL-6、TNF、VEGFA等靶点及TNF信号通路、PI3K-Akt信号通路、HIF-1信号通路等多靶点多通路发挥治疗作用。
Objective:To study the medication rule and mechanism of core drug action of traditional Chinese medicine compound patent for the treatment of heart failure in China National Intellectual Property Administration database.Methods:The patent retrieval and analysis system of the China National Intellectual Property Administration was searched.The patents of chinese herbal compound for the treatment of heart failure from January 2002to February2021were collected,and the database was established.The statistics of usage frequency,medicinal properties,flavor of medicinals,meridian tropism,efficacy classification and syndrome type of traditional Chinese medicines were made.Aprirori algorithm correlation and medicine relevance network of commonly used drugs were analyzed,and the core drugs for the treatment of heart failure were analyzed.The TCMSP database was used to retrieve the active components and action targets of high-frequency core drugs.The CTD,OMIM and GeneCardes databases were used to retrieve disease-related targets.The STRING database was used to construct the protein-protein interaction network.The David database was used to perform GO analysis and KEGG signaling pathway analysis.The Cytoscape was used to construct the component-target-pathway network.Results:A total of 161patents of chinese herbal compound for the treatment of heart failure were collected,including 423TCM,78of which had frequency≥5and 18of which had frequency≥21,10high-frequency couplet medicines and 10high-frequency angle medicines were obtained.The classification of medicines mainly includes qi-tonifying medicinal,blood-activating medicinal,damp-draining diuretic medicinal and interior-warming medicinal,and the core herbs were Radix Astragali,Radix Salvia Miltiorrhiza,Poria,and Radix Aconiti Lateralis Preparata.Network pharmacological analysis showed that it mainly acted on IL-6,TNF,VEGFA,PTGS2,NOS2and other potential targets in the treatment of heart failure,regulating TNF signaling pathway,PI3K-Akt signaling pathway,HIF-1signaling pathway and so on.Conclusion:The traditional Chinese medicine compound patent for the treatment of heart failure mainly combined qi-tonifying medicinal with blood-activating medicinal,and combined damp-draining diuretic medicinal with interior-warming medicinal.The core herbs are Radix Astragali,Radix Salvia Miltiorrhiza,Poria,and Radix Aconiti Lateralis Preparata.These core herbs may treat heart failure by intervening IL-6,TNF,VEGFA and other targets and regulating through TNF signaling pathway,PI3K-Akt signaling pathway,HIF-1signaling pathway and other pathways.
