摘要
目的 研究长链非编码RNA铁蛋白重链1伪基因3(LncRNA FTH1P3)在胃癌组织中的表达及临床意义,并探讨LncRNA FTH1P3通过调控磷脂酰肌醇3-激酶(PI3K)/苏氨酸蛋白激酶(AKT)信号通路促进胃癌细胞增殖和迁移的作用机制。方法 收集胃癌和癌旁组织标本,采用qRT-PCR检测LncRNA FTH1P3在胃癌组织中的表达,分析LncRNA FTH1P3的表达与胃癌患者病理参数和预后的关系。常规培养胃癌细胞,分为si-NC组和si-LncRNA FTH1P3-1组、si-LncRNA FTH1P3-2组,采用LncRNA FTH1P3 siRNA转染胃癌细胞,qRT-PCR检测各组细胞中LncRNA FTH1P3的表达;CCK8实验检测各组细胞增殖能力;Transwell实验检测各组细胞迁移能力;体内成瘤实验检测各组细胞体内成瘤能力;Western blot检测各组细胞PI3K/AKT信号通路相关蛋白PI3K、AKT、磷酸化PI3K(pPI3K)、磷酸化AKT(pAKT)的表达。结果 LncRNA FTH1P3在胃癌组织中表达上调(P<0.05)。LncRNA FTH1P3高表达与患者肿瘤最大径、分化程度、淋巴结转移情况和TNM分期有关(P<0.05)。与si-NC组相比,si-LncRNA FTH1P3-1组和si-LncRNA FTH1P3-2组胃癌细胞中LncRNA FTH1P3表达显著减少(P<0.05),si-LncRNA FTH1P3-1组和si-LncRNA FTH1P3-2组胃癌细胞增殖、迁移、体内成瘤能力显著降低(P<0.05);细胞中pPI3K和pAKT的表达减少(P<0.05)。结论 LncRNA FTH1P3通过调控PI3K/AKT信号通路促进胃癌细胞增殖及迁移,有望成为治疗胃癌的潜在分子靶点。
Objective To study the expression level and clinical significance of long noncoding RNA ferritin heavy chain 1 pseudogene 3(LncRNA FTH1 P3) in gastric cancer tissues, and to investigate the mechanism of LncRNA FTH1 P3 to regulate phosphatidylinositol-3-kinase(PI3 K)/AKT signaling pathway to promote the proliferation and migration of gastric cancer cells.Methods Gastric cancer and adjacent tissue samples were collected, qRT-PCR was used to detect the expression level of LncRNA FTH1 P3 in gastric cancer tissues, and analyzed the relationship between the expression of LncRNA FTH1 P3 and the pathological parameters and prognosis of patients with gastric cancer.Routinely cultured gastric cancer cells and they were divided into si-NC group, si-LncRNA FTH1 P3-1 group, si-LncRNA FTH1 P3-2 group, gastric cancer cells were transfected with LncRNA FTH1 P3 siRNA,and the expression level of LncRNA FTH1 P3 in each group was detected by qRT-PCR;CCK8 experiment was used to detect cell proliferation ability of each group;Transwell experiment was used to detect cell migration ability of each group;in vivo tumor formation experiment was used to detect tumor formation ability of each group;Western blot was used to detect the expression of PI3 K/AKT signaling pathway related proteins PI3 K,AKT,phosphorylated-PI3 K(pPI3 K),phosphorylated-AKT(pAKT) of each group.Results LncRNA FTH1 P3 expression was up-regulated in gastric cancer tissues(P<0.05).The high expression of LncRNA FTH1 P3 was correlated with the maximum tumor diameter, tumor differentiation, lymph node metastasis, and TNM staging(P<0.05).Compared with the si-NC group, the expression of LncRNA FTH1 P3 in gastric cancer cells of the si-LncRNA FTH1 P3-1 group and the si-LncRNA FTH1 P3-2 group were significantly reduced(P<0.05),and the proliferation, migration and tumor formation ability of the si-LncRNA FTH1 P3-1 group and si-LncRNA FTH1 P3-2 gastric cancer cells were significantly reduced(P<0.05);the expression of pPI3 K and pAKT in the cells were reduced(P<0.05).Conclusion LncRNA FTH1 P3 regulates the PI3 K/AKT signaling pathway to promote the proliferation and migration of gastric cancer cells, which may be a potential molecular target for the treatment of gastric cancer.
