摘要
目的建立同时测定醒脾养儿颗粒中新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C 6种咖啡酰奎宁酸类成分含量的高效液相色谱法。方法色谱柱为ACE Excel 5 C柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸水溶液(梯度洗脱),流速为1.0 mL/min,检测波长为325 nm,柱温为30℃,进样量为10μL。结果新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C质量浓度分别在2.65~53.06μg/mL、4.20~84.00μg/mL、3.43~68.57μg/mL、1.40~27.96μg/mL、0.72~14.47μg/mL、2.06~41.45μg/mL范围内与峰面积线性关系良好(r≥0.9996,n=6);精密度、稳定性、重复性试验结果的RSD均低于2.0%(n=6);平均加样回收率分别为100.48%,99.34%,102.95%,103.13%,102.03%,103.02%,RSD分别为1.09%,0.73%,1.71%,2.36%,2.43%,0.53%(n=6)。21批样品中6种咖啡酰奎宁酸类成分的总含量为0.30291~0.68319 mg/g,平均含量为0.48599 mg/g。结论该方法操作简便、重复性好,可用于醒脾养儿颗粒中6种咖啡酰奎宁酸类成分的含量测定。
Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of six caffeoylquinic acids including neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in Xingpi Yang’er Granules.Methods The chromatographic column was ACE Excel 5 Ccolumn(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.2%phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 325 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C were 2.65-53.06μg/mL,4.20-84.00μg/mL,3.43-68.57μg/mL,1.40-27.96μg/mL,0.72-14.47μg/mL and 2.06-41.45μg/mL(r≥0.9996,n=6),respectively.The RSDs of precision,stability and repeatability tests were lower than 2.0%(n=6).The average recoveries of the above six caffeoylquinic acids were 100.48%,99.34%,102.95%,103.13%,102.03%and 103.02%with RSDs of 1.09%,0.73%,1.71%,2.36%,2.43%,and 0.53%(n=6),respectively.The total content of six caffeoylquinic acids in the 21 batches of samples was in the range of0.30291-0.68319 mg/g,with the average content of 0.48599 mg/g.Conclusion The method is simple and repeatable,which can be used for the determination of six caffeoylquinic acids in Xingpi Yang’er Granules.
