摘要
采集了新疆南疆地区骏枣缩果病的病样,通过对病样的分离培养、致病性测定、形态特征和培养性状观察,并结合18 S rDNA-ITS、EF-1α、β-tubulin、ATPase多基因序列进行了系统发育分析。结果表明:经分离培养和致病性验证,确定了新疆南疆骏枣缩果病的病原,采用18 S rDNA-ITS、EF-1α和ATPase无法有效区分链格孢菌Alternaria alternate和细极链格孢菌Alternaria tenuissima。通过β-tubulin基因序列、核苷酸一致性进行比较分析和形态学鉴定,将新疆南疆骏枣缩果病病原鉴定为链格孢菌Alternaria alternate(Fr.)Keissler。
The samples were collected from Southern Xinjiang.The phylogenetic analysis were carried out based on the isolation and culture,pathogenicity test,Morphology,cultural characteristics and the 18 S rDNA-ITS,EF-1αandβ-tubulin gene sequence analysis.The results showed that the pathogen of Jun jujube fruit shrink disease was determined by isolating,culturing,and pathogenicity test.The 18 S rDNA-ITS,EF-1αand ATPase gene sequence could not effectively distinguish Alternaria alternate from Alternaria tenuissima.According to the morphological features of culture,theβ-tubulin gene sequence and nucleotide identity analysis of the strains,the pathogen was identified as Alternaria alternata(Fr.)Keissler.
