摘要
目的:探究新对叶百部碱对肺炎支原体诱导的肺炎支原体肺炎小鼠致咳因子TRPA1(瞬时受体电位离子通道蛋白)表达的影响。方法:将50只雄性C57小鼠随机分为正常对照组、模型组、新对叶百部碱组、阿奇霉素组、枸橼酸喷托维林组;除正常组外(予以鼻腔滴注等量生理盐水),其余5组小鼠鼻腔滴入50μL培养完成的肺炎支原体液制备肺炎支原体肺炎模型,连续给药7 d后,采用氨水引咳法,对5组小鼠进行引咳实验,记录咳嗽次数及咳嗽潜伏期。采用HE染色观察肺组织病理变化,免疫组织化学法观察肺内TRPA1蛋白表达,RT-PCR技术检测小鼠肺组织TRPA1 mRNA表达。结果:引咳实验结果显示新对叶百部碱组与模型组相比较咳嗽次数及咳嗽潜伏期均有改善,HE染色显示给药后感染小鼠肺组织结构有所改善。免疫组化结果显示,与模型组相比,新对叶百部碱组TRPA1蛋白表达减弱;RT-PCR结果显示;新对叶百部碱在一定程度上能降低TRPA1 mRNA表达。结论:新对叶百部碱可以通过抑制肺组织TRPA1表达发挥止咳镇咳作用。
Objective:To investigate the effect of Nephrine on the expression of TRPA1(transient receptor potential ion channel protein)in mice with mycoplasma pneumoniae pneumonia induced by mycoplasma pneumoniae.Methods:50 male C57 mice were randomly divided into normal control group,model group,Neotuberostemonine group,Azithromycin group and Pentoverine citrate group with each 10 mice.Except for the normal group(the same amount of normal saline nasal instillation),the other 5 groups of mice were given 50μL cultured mycoplasma pneumoniae body fluid for nasal instillation to prepare mycoplasma pneumoniae pneumonia model.After 7 days of continuous administration,the mice in the 5 groups were subjected to cough induction test by ammonia water,and The Times of cough and the incubation period of cough were recorded.The pathological changes of lung tissues were observed by HE staining,the expression of TRPA1 protein in lung tissues was observed by immunohistochemistry,and the expression of TRPA1 mRNA in lung tissues of mice was detected by RT-PCR.Results:The result of cough-inducing experiment showed that compared with the model group,the number of coughs and the incubation period of cough in the Neotuberostemonine group were improved,and HE staining showed that the lung tissue structure of infected mice in Neotuberostemonine group was improved after administration.Immunohistochemical results showed that compared with the model group,the expression of TRPA1 protein in the Neotuberostemonine group was weakened.RT-PCR results showed that the TRPA1 mRNA expression in the Neotuberostemonine group was reduced to some extent.Conclusion:The Neotuberostemonine can play a role in relieving cough and cough by inhibiting TRPA1 expression in lung.
