摘要
目的研究依达拉奉右莰醇治疗血管性痴呆大鼠的疗效及作用机制。方法双侧颈总动脉永久结扎术(2VO)造模后,30只大鼠随机分至对照组、模型组和依达拉奉右莰醇治疗组。治疗组给予依达拉奉右莰醇1.25 mg/kg腹腔注射每12小时(H)1次,其余组同方法等量0.9%氯化钠溶液治疗,疗程2周。Morris水迷宫法检测大鼠行为学,免疫组化染色观察和测定海马组织半胱氨酸天冬氨酸蛋白水解酶-3(Caspase-3)、β淀粉样蛋白(Aβ)的含量。结果与模型组比较,依达拉奉右莰醇治疗后VD大鼠穿台次数[3.5(3,4)]增加,逃避潜伏期(36.56±2.86)s缩短,海马区Aβ/Caspase-3平均光密度(AOD)值(0.082±0.002/0.076±0.002)减低,差异有统计学意义(P<0.05)。结论依达拉奉右坎醇可能通过下调海马区凋亡蛋白的表达来使VD大鼠在学习记忆上有所恢复。
Objective To study the efficacy and mechanism of Edaravone Dexborneol in the treatment of rats with vascular dementia(VD).Methods Thirty rats were randomly divided into the control group,model group and Edaravone Dexborneol treatment group by modeling with permanent ligation of bilateral common carotid arteries occlusion(2VO).The rats in the treatment group were administered with peritoneal injection of Edaravone dexborneol 1.25mg/kg(q.12h),and rats in the remaining groups only received normal saline in equivalent volume by the same method.2 weeks of treatment were conducted,Morris water maze was used to examine the behavior of rats,and immunohistochemical staining was performed to observe and measure the contents of Cysteinyl aspartate specific proteinase 3(Caspase-3)and beta amyloid(Aβ)protein in hippocampus.Results Compared with the model group,the frequency of cross-platform times was increased to 3.5(3,4),and the escape latency was shortened to(36.56±2.86),and the average optical density(AOD)of Aβ/Caspase-3 in the hippocampus in the treatment group was decreased to(0.082±0.002/0.076±0.002),and the differences were statistically significant between groups(P<0.05).Conclusion Edaravone dextrocanol supports VD rats learn and remember better by down-regulating apoptotic proteins in the hippocampus.
