菊花CmSDHs基因的克隆和功能分析

摘要 本研究通过生物信息学分析方法,对CmSDH2和CmSDH6基因氨基酸序列进行结构、疏水性和亲水性分析,为解析该基因的功能和在弱光条件下的响应规律提供理论依据。根据菊花转录组中琥珀酸脱氢酶基因Cm SDH2和CmSDH6序列信息进行同源克隆,分别获得579 bp和423 bp的基因全长,以‘南农雪峰’和‘南农宫粉’为试验材料,利用同源克隆方法从转录组中克隆出Cm SDH2和Cm SDH6基因,设置80%自然光照(对照)和20%自然光照(弱光处理),进行差异表达分析。结果表明:Cm SDH2和Cm SDH6基因的相对表达量在弱光条件显著下降,因此推断该基因在抗弱光过程中起到负调控作用。 This study uses bioinformatics analysis methods to analyze the structure,hydrophobicity and hydrophilicity of the amino acid sequences of CmSDH2 and Cm SDH6 genes to provide a theoretical basis for analyzing the function of the gene and the response law under low light conditions.According to the sequence information of the succinate dehydrogenase genes CmSDH2 and CmSDH6 in the chrysanthemum transcriptome,homologous cloning was performed,and the gene of 579 bp and 423 bp were obtained,respectively.The test materials were’Nannong Xuefeng’and’Nannong Gongfen’CmSDH2 and CmSDH6 genes which were cloned from the transcriptome using the homologous cloning method,and 80%natural light(control)and 20%natural light(low light treatment)were set for differential expression analysis.The results showed that the relative expression of CmSDH2 and CmSDH6 genes decreased significantly under low light conditions,it is inferred that the genes play a negative regulatory role in the process of low light resistance.
