中药过敏康方对银屑病小鼠模型皮损及免疫因子的调节作用

Regulatory Effect of Traditional Chinese Formula Guominkang(过敏康)on Skin Lesion and Immunologic Factors in Mice with Psoriasis

摘要 目的研究中药过敏康方对银屑病小鼠模型皮损组织及免疫因子的调节作用,评价中药过敏康方(Guominkang Decoction,GMK)干预银屑病小鼠模型的作用及机制。方法将50只Balb/c雄性小鼠分为5组,分别为正常组(生理盐水)、模型组(咪喹莫特)、甲氨蝶呤组(咪喹莫特+1 mg/kg)、过敏康方高剂量组[咪喹莫特+5.6 g/kg,GMK(H)]、过敏康方低剂量组[咪喹莫特+2.8 g/kg,GMK(L)],每日涂抹咪喹莫特、灌胃给药各1次,连续给药6 d。给药开始每天对各组小鼠进行表皮厚度及银屑病皮损面积和严重程度指数(PASI)评分;实验结束后,剪取小鼠造模皮肤进行形态学、组织病理学观察;并用酶联免疫吸附试验法(ELISA)检测各组小鼠皮损组织中免疫因子白细胞介素(Interleukin,IL)-6、IL-1β、IL-23、IL-8、IL-10、IL-22、IL-17A的表达。结果与正常组相比,模型组皮损部位的鳞屑较多,出现红斑,皮肤厚度显著性增高(P<0.001);皮损组织中IL-6、IL-1β、IL-23、IL-8、IL-22、IL-17A显著性升高(P<0.001);与模型组相比,各给药组皮损部位的鳞屑、皮肤厚度等具有明显的改善;病理学观察皮损组织切片,各给药组均变薄,具有显著性差异(P<0.001);皮损组织中IL-6、IL-1β、IL-23、IL-8、IL-22、IL-17A,各给药组均具有显著性降低。结论过敏康方高、低剂量组均具有抑制银屑病小鼠模型的作用,可能与降低IL-6、IL1β、IL-23、IL-8、IL-22、IL-17A,升高IL-10相关。 Objective To study the regulation of the traditional Chinese medicine formula Guominkang(过敏康,GMK)on the skin lesions and immune factors of psoriasis mouse models,and to evaluate the effect and mechanism of GMK on psoriatic mouse model.Methods 50 Balb/C male mice were divided into 5 groups,which were control group(glycerol),model group(Imiquimod),methotrexate group(Imiquimod and 1 mg/kg),GMK with high-dose group[Imiquimod and 5.6 g/kg,GMK(H)],and low-dose group[Imiquimod and 2.8 g/kg,GMK(L)],one time of every day,six consecutive days.At the beginning,the skin thickness and PASI score of mice in each group were measured every day.After the treatment,the model skin was cut off for morphological and histopathological observation,and the expression of immunologic factors(IL-6,IL-1β,IL-23,IL-8,IL-10,IL-22,IL-17A)on skin lesion were detected by Elisa.Results Compared with the normal group,the model group had more scales,erythema,and a significant increase in skin thickness(P<0.001);IL-6,IL1β,IL-23,IL-8,IL-22,and IL-17A in the skin lesions increased significantly(P<0.001);the epidermal thickness of each treatment group was thinner than that of the model group(P<0.001),and the high dose group had a statistically significant decrease compared with the positive control group(P<0.001);the expression of immunologic factors(IL-6,IL-1β,IL-23,IL-8,IL-22,IL17A)on skin lesion of the treatment groups were lower than those of the model group,and the high dose group was obviously lower than the positive control group on the expression of IL-6 and IL-1β.Conclusion After external administration of different doses of GMK,the skin lesions of psoriasis mice model can be improved in varying degrees,which may be related to the regulation of immunologic factors(IL-6,IL-1β,IL-23,IL-8,IL-22,IL-10,IL-17A)in skin lesions.
