黄梁木枝叶水提物的抗菌作用及对小鼠免疫和肠道菌群的影响 被引量:1

Effects of aqueous extracts from branches and leaves of Neolamarckia cadamba(Roxb.)Bosser on biochemical indices and intestinal flora of mice

摘要 黄粱木(Neolamarckia cadamba(Roxb.)Bosser)具有多种药理作用,但发挥作用的活性成分及作用机理仍有待揭示,本研究旨在评价黄梁木枝叶水提物对小鼠抗炎、抗氧化作用及对肠道菌群的影响。采用热水浸提法获得黄梁木枝叶水提物(aqueous extract of Neolamarckia cadamba branch and leaf, NCAE)并进行体外抑菌活性检测。将24只小鼠随机分为空白组、黄粱木组(3 g水提物/kg)和抗生素组(0.01 g/kg庆大霉素),每组8只,进行动物试验。采用试剂盒法测定小鼠过氧化氢酶(CAT)、丙二醛(MDA)、超氧化物歧化酶(SOD)、IL-2、IL-6、TNF-α等因子含量,高通量测序法分析小鼠肠道菌群变化。结果显示,NCAE中含有多糖、黄酮、总酚和三萜皂苷等活性物质,且对金黄色葡萄球菌、猪大肠杆菌、猪霍乱沙门菌均显示抑菌活性。活体试验表明,黄粱木组小鼠血清、脾脏和肾脏中IL-6含量显著下降(P<0.05);血清和肾脏中TNF-α含量显著降低(P<0.05);黄粱木组小鼠脾脏和肾脏中CAT含量显著升高(P<0.05);而血清、脾脏和肾脏中MDA含量显著降低(P<0.05);饲喂NCAE后小鼠盲肠中另枝菌属、瘤胃球菌属的含量显著升高(P<0.05),毛螺菌属、拟普雷沃菌属、罗氏菌属和厌氧棍状菌属的含量显著降低(P<0.05)。结果表明,NCAE具有体外抑菌活性,在体内有提高机体抗炎和抗氧化能力,增加盲肠微生物菌群丰富度、调节菌群种类和数量,具有替代抗生素的潜力。 Neolamarckia cadamba(Roxb.) Bosser has a variety of pharmacological effects, but the active components and their mechanism are still unknown.The purpose of this study was to evaluate the antibacterial, anti-inflammatory and antioxidant effects of water extracts from branches and leaves of Neolamarckia cadamba(Roxb.) Bosser,and its effects on the intestinal flora of mice.Aqueous extract of Neolamarckia cadamba branches and leaves(NCAE) was prepared by hot water extraction, and its antibacterial activity was detected in vitro.Then 24 mice were randomly divided into blank group, NCAE group and antibiotic group, with 8 mice in each group.After the experiment, the contents of CAT,MDA,SOD,IL-2,IL-6,TNF-α and other factors were determined by ELISA kit, and the changes of intestinal flora were analyzed by high-throughput sequencing method.The results showed that NCAE contained polysaccharides, flavonoids, total phenols and triterpenoid saponins, and showed antibacterial activities against Staphylococcus aureus,Escherichia coli and Salmonella cholerae.The in vivo test showed that the contents of IL-6in serum,spleen and kidney of mice were significantly decreased in NCAE group(P<0.05),and the contents of TNF-αin serum and kidney were also significantly decreased(P<0.05).CAT content in spleen and kidney of NCAE group was significantly increased(P<0.05),while the content of MDA in serum,spleen and kidney was significantly decreased(P<0.05)in NCAE group.NCAE feeding significantly increased the contents of Alistipes and Ruminococcus in caecum of mice(P<0.05),while significantly decreased the contents of Lachnospiraceae,Alloprevotella,Roseburia and Anaerotruncus(P<0.05).The results of this study indicate that NCAE has antibacterial activity in vitro and can not only improve the anti-inflammatory and antioxidant capacity of the body in vivo,but also increase the richness of cecal microflora and regulate the species and quantity of microflora,which has the potential to replace antibiotics.
