摘要
目的:建立藏药吉尼德协化学成分的超高效液相色谱-四级杆/静电场轨道阱高分辨质谱(UPLC-Q-Exactive Orbitrap MS)定性分析方法。方法:采用ZORBAX SB-C_(18)色谱柱(150 mm×2.1 mm,1.8μm)进行分离,以甲醇-0.1%甲酸溶液为流动相进行梯度洗脱;在正、负离子监测模式下采集数据。采用Xcalibur 2.0软件计算出化合物的精确分子量、准分子离子峰、特征碎片离子等信息,并参考ChemSpider,mzCloud和mzVault质谱数据库及文献报道对吉尼德协的化学成分进行识别和鉴定。结果:共鉴定出85种化学成分,包括12种生物碱类、18种黄酮类、20种有机酸类、19种酚类、9种萜类及7种其他类别成分。结论:本研究首次对吉尼德协的化学成分进行全面定性表征,为其进一步的药效物质基础研究和质量控制提供了科学依据。
Objective:To establish a method for the qualitative analysis of the chemical components of Tibetan medicine Ji-ni-de-xie by ultra high performance liquid chromatography with four stage rod/electrostatic field orbital trap high resolution mass spectrometry(UPLC-Q-Exactive Orbitrap MS).Methods:ZORBAX SB-C_(18) column(150 mm×2.1 mm,1.8μm)was used for separation,and methanol-0.1%formic acid solution was used as the mobile phase for gradient elution.Data was collected in positive and negative ion monitoring modes.Xcalibur 2.0 software was used to calculate the precise relative molecular mass,quasi-molecular ion peak,and characteristic fragment ions of the compounds.In addition,ChemSpider,mzCloud and mzVault databases and literature were referred to to identify the chemical composition of Ji-ni-de-xie.Results:A total of 85 chemical components were identified,including 12 alkaloids,18 flavonoids,20 organic acids,19 phenols,9 terpenoids,and 7 other components.Conclusion:This study conducted a comprehensive and qualitative characterization of the chemical components of Ji-ni-de-xie for the first time,which provides a scientific basis for its further effective substances research and quality control.
