四妙衍生方对高尿酸模型大鼠肠道菌群的影响 被引量:3

摘要 目的研究四妙衍生方对高尿酸模型大鼠肠道菌群的影响。方法将24只雄性SD大鼠分为正常对照组(等体积生理盐水)、模型组(等体积生理盐水)和四妙衍生方组(生药30 g/kg),各8只;喂饲高嘌呤饲料(含次黄嘌呤100 g/kg),隔日1次皮下注射氧嗪酸钾(200 mg/kg),每日1次10~12℃冷水浴10 min,连续6 d,以复制高尿酸大鼠模型,正常对照组予常规饲养;建模同时,各组大鼠灌胃相应药物或生理盐水,每日1次,连续6 d。测定大鼠血清尿酸和尿素氮水平,进行16S rRNA测序并分析大鼠直肠部位粪便菌群变化情况。结果与模型组比较,四妙衍生方组大鼠血清尿酸、尿素氮水平均显著降低(P<0.01),拟杆菌门、瘤胃球菌属_UCG-014、肠球菌属、乳杆菌属丰度均显著降低(P<0.01),Allobaculum、杜氏杆菌属、瘤胃球菌属_NK4A214、罗姆布茨菌属丰度均显著升高(P<0.05或P<0.01)。结论四妙衍生方可降低高尿酸模型大鼠尿酸水平、改善其肠道菌群丰度。通过肠道菌群变化直接调控尿酸水平或可成为抗高尿酸中成药的研发新思路。 Objective To study the effect of Simiao Derivative Formula(SDF)on intestinal microflora in rats with hyperuricemia.Methods Twenty-four male SD rats were divided into the normal control group(equal volume of normal saline),model group(equal volume of normal saline)and SDF group(equivalent to 30 g/kg of crude drug),eight rats in each group.The rats were fed with high purine diet(including 100 g/kg of hypoxanthine),subcutaneously injected with potassium oxalate(200 mg/kg)once every other day,and bathed in cold water at 10-12℃for 10 min once a day for 6 d to duplicate the model rat with hyperuricemia.The rats in the normal control group were routinely fed.At the same time of modeling,rats in each group were perfused with corresponding drugs or normal saline once a day for 6 d.The levels of uric acid and urea nitrogen in the serum of rats were measured,the 16S rRNA was sequenced,and fecal microflora in the rectum of rats was analyzed.Results Compared with those in the model group,the levels of serum uric acid and urea nitrogen were significantly lower(P<0.01),the abundance of Bacteroides,Ruminococcaceae_UGG-014,Enterococcus and Lactobacillus was significantly lower(P<0.01),while the abundance of Allobaculum,Dubosiella,Ruminococcaceae_NK4A214 and Rumboutsia was significantly higher in the SDF group(P<0.05 or P<0.01).Conclusion SDF can reduce the level of uric acid and improve the abundance of intestinal flora in rats with hyperuricemia.The direct regulation of the uric acid level through the change of intestinal microflora may become a new idea for the research and development of anti-hyperuricemic Chinese patent medicine.
