血清miR-137、miR-335表达与妊娠期糖尿病相关性分析

摘要 目的探究血清微小RNA-137(miR-137)、微小RNA-335(miR-335)表达与妊娠期糖尿病(GDM)的相关性。方法采用回顾性研究,选取2017年6月至2019年12月于海南医学院第一附属医院就诊的GDM孕妇96例纳入观察组及产检的健康孕妇96例作为对照组。检测两组孕妇血清高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、血管细胞黏附分子-1(VCAM-1)、miR-137、miR-335水平,并进行Pearson相关性分析和Logistic回归分析。结果观察组血清LDL-C、VCAM-1、miR-137水平为(3.45±0.39)mmol/L、(110.65±29.76)ng/mL、1.72±0.51,均显著高于对照组[(2.62±0.28)mmol/L、(61.57±18.62)ng/mL、1.02±0.27],HDL-C、miR-335水平为(0.94±0.21)mmol/L、0.62±0.16,均低于对照组[(1.08±0.27)mmol/L、1.08±0.29],差异均有统计学意义(P<0.05)。Pearson相关性分析结果显示,血清miR-137水平与LDL-C、VCAM-1呈正相关,与HDL-C呈负相关(P<0.05);血清miR-335与LDL-C、VCAM-1呈负相关,与HDL-C呈正相关(P<0.05);多因素Logistic回归分析结果显示,血清VCAM-1、miR-137及miR-335水平均为影响孕妇发生GDM的独立危险因素(P<0.05)。结论GDM患者血清miR-137表达水平升高,血清miR-335表达水平降低,二者可能参与GDM发生、发展,并可能与血管内皮细胞损伤及脂质代谢异常有关。 Objective To explore the relationships between the expressions of microRNA-137(miR-137),microRNA-335(miR-335)and gestational diabetes mellitus(GDM).Methods Using a retrospective study,a total of 96 GDM pregnant women who visited the First Affiliated Hospital of Hainan Medical University from June 2017 to December 2019 were included in the observation group and 96 healthy pregnant women as control group.The levels of serum low density lipoprotein cholesterol(LDL-C),high density lipoproteinajiuxen cholesterol(HDL-C),vascular cell adhesion molecule-1(VCAM-1),miR-137,miR-335 were measured,and Pearson correlation analysis and Logistic regression analysis were performed.Results The levels of LDL-C,VCAM-1 and miR-137 in the observation group were(3.45±0.39)mmol/L,(110.65±29.76)ng/mL,1.72±0.51,which were significantly higher than those in the control group[(2.62±0.28)mmol/L,(61.57±18.62)ng/mL,1.02±0.27],and the levels of HDL-C and miR-335 were(0.94±0.21)mmol/L,0.62±0.16,which were significantly lower than those in the control group[(1.08±0.27)mmol/L,1.08±0.29],the differences were statistically significant(P<0.05).Pearson correlation analysis showed that serum miR-137 level was positively correlated with LDL-C and VCAM-1,and negatively correlated with HDL-C(P<0.05);serum miR-335 was negatively correlated with LDL-C and VCAM-1,and positively correlated with HDL-C(P<0.05);multivariate Logistic regression analysis showed that the levels of serum VCAM-1,miR-137 and miR-335 were independent risk factors for GDM in pregnant women(P<0.05).Conclusion The expression level of serum miR-137 in GDM patients is increased,and the expression level of serum miR-335 is decreased,which may be involved in the occurrence and development of GDM,and may be related to the injury of vascular endothelial cells and the abnormality of lipid metabolism.
