人成纤维细胞生长因子受体1基因表达抑制通过线粒体途径影响血管瘤内皮细胞凋亡的研究

Research of human fibroblast growth factor receptor 1 gene expression inhibition affects hemangioma endothelial cells apoptosis through the mitochondrial pathway

摘要 目的探讨人成纤维细胞生长因子受体1(fibroblast growth factor receptor 1,FGFR1)基因表达抑制通过线粒体途径影响血管瘤内皮细胞(hemangioendo⁃thelial cells,HemECs)凋亡。方法将HemECs分为空白组(仅加入脂质体)、对照组(转染阴性对照siRNA)和实验组(转染靶向抑制FGFR1的siRNA)。转染48 h,用流式细胞术检测细胞凋亡率,用蛋白质印迹法检测FGFR1、B淋巴细胞瘤-2相关X蛋白(B lymphoma-2 related X protein,Bax)、B细胞淋巴瘤/白血病-X基因长片段(B cell lymphoma/leukemia-X gene long fragment,Bcl-xl)、半胱氨酸天冬氨酸蛋白水解酶3(cysteine aspartate proteolytic enzyme 3,Caspase-3)和Caspase-9蛋白的表达水平。结果干预细胞48 h,实验组、对照组和空白组的细胞凋亡率分别为(19.27±1.67)%,(3.47±0.21)%和(3.62±0.24)%,FGFR1蛋白相对表达量分别为0.07±0.01,0.56±0.05和0.53±0.04,Bax蛋白相对表达量分别为0.35±0.03,0.12±0.02和0.11±0.01,Bcl-xL蛋白相对表达量分别为0.13±0.02,0.50±0.05和0.49±0.05,Caspase-3相对表达量分别为0.37±0.04,0.12±0.01和0.12±0.01,Caspase-9相对表达量分别为0.26±0.03,0.10±0.01和0.10±0.01,实验组的上述指标与对照组和空白组比较,差异均有统计学意义(均P<0.05)。结论抑制FGFR1基因表达可通过线粒体途径影响HemECs凋亡。 Objective To explore the human fibroblast growth factor receptor 1(FGFR1)gene expression inhibition affects the apoptosis of hemangioendothelial cells(HemECs)through the mitochondrial pathway.Methods HemECs were divided into blank group(only liposomes were added),control group(transfected with negative control siRNA)and experimental group(transfected with siRNA targeting FGFR1).After transfection for 48 hours,the apoptosis rate was detected by flow cytometry,the expression levels of FGFR1,B lymphoma-2 associated X protein(Bax),B cell lymphoma/leukemia-X gene long fragment(Bcl-xl),cysteine aspartate proteolytic enzyme 3(Caspase-3)and Caspase-9 protein were detected by Western blot.Results Intervention cells for 48 h,the apoptosis rates of the experimental,control and blank groups were(19.27±1.67)%,(3.47±0.21)%and(3.62±0.24)%,the relative expression levels of FGFR1 protein were 0.07±0.01,0.56±0.05 and 0.53±0.04,the relative expression levels of Bax protein were 0.35±0.03,0.12±0.02 and 0.11±0.01,the relative expression levels of Bcl-xL protein were 0.13±0.02,0.50±0.05 and 0.49±0.05,the relative expression levels of Caspase-3 were 0.37±0.04,0.12±0.01 and 0.12±0.01,the relative expression levels of Caspase-9 were 0.26±0.03,0.10±0.01 and 0.10±0.01.The above indicators of experimental group were compared with those of the control and blank groups,the differences were statistically significant(all P<0.05).Conclusion Inhibition of GFR1 gene expression can affect HemECs apoptosis through the mitochondrial pathway.
