摘要
目的:运用网络药理学方法,分析经典方剂茵陈蒿汤(YCHD)治疗非酒精性脂肪肝病(NAFLD)的作用机制。方法:在TCMSP和GeneCards、OMIM等数据库分别检索YCHD中各中药的主要化学成分、作用靶点和NAFLD的疾病靶点。在STRING和Metascape平台分别分析PPI网络和其参与的生物过程和相关通路。然后利用Cytoscape V3.8.2软件构建“YCHD成分-NAFLD靶点”网络。最后通过分子对接分析核心成分与核心靶标的结合情况。结果:筛选得到29种YCHD治疗NAFLD的活性成分和180个相关作用靶点;YCHD治疗NAFLD的生物学通路主要富集在AGE-RAGE信号通路、动脉粥样硬化等,其功能主要为调节转录因子结合、细胞因子受体等活性的生命活动。分子对接的结果表明核心成分与核心靶点结合能较低,结合活性较好。结论:本研究初步阐释了YCHD治疗NAFLD的多成分、多靶点、多途径的作用机制,为YCHD的临床推广应用及药物开发提供了理论研究基础。
Objective:To analyze the mechanism of Yinchenhao Decoction(YCHD)in treating non-alcoholic fatty liver disease based on network pharmacology.Methods:The main chemical components,action targets and disease targets of NAFLD of each herbal medicine in YCHD were searched in TCMSP and GeneCards,OMIM and other databases,respectively.PPI networks and their involvement in biological processes and related pathways were analyzed in STRING and Metascape platforms,respectively.Then Cytoscape v3.8.2 software was used to construct the“YCHD component-NAFLD target”network.Finally,the binding of the core components to the core targets was analyzed by molecular docking.Results:Through screening,29 active ingredients of YCHD for the treatment of NAFLD and 180 relevant action targets were obtained;the biological pathway of YCHD for NAFLD was mainly enriched in AGE-RAGE signaling pathway,atherosclerosis,etc.Its function was mainly to regulate the vital activities of transcription factor binding,cytokine receptors and other activities.The results of molecular docking showed low binding energy and good binding activity of the core components to the core targets.Conclusion:This study initially elucidate the multi-component,multi-target and multi-pathway mechanism of action of YCHD in the treatment of NAFLD,providing a theoretical research basis for the clinical promotion and application of YCHD and drug development.
