摘要
[目的]探究水处理周期中微生物群落的动态变化情况。[方法]采用EPS泡沫滤珠作为生物滤料开展水处理试验,通过水质指标氨氮、亚硝酸盐、TOC的测定来评估其水质处理效果,另采用Illumina miseq高通量进行水体中细菌的16S rRNA基因V3+V4区的测序,通过分析微生物群落结构及多样性来探究整个水处理周期中的微生物群落结构变化。[结果]在10 d的水处理过程中,氨氮、亚硝酸盐、TOC的最终去除率分别可达98.0%、99.0%和95.0%以上,在第5天时水质情况趋于稳定。分析不同时间水体中细菌丰度及多样性的变化发现,水体中的微生物丰富度在第9天最高,群落多样性在第5天最高。整个周期中优势细菌在门分类上变化不大,变形菌门在水体中占主要优势,在前3 d占比达95.00%以上,第5天之后从77.79%逐渐下降至63.57%;在属分类层面上,水体中细菌丰富度属类差异较大,其中弧菌属在第1天占比为2.97%,第3天迅速增殖至98.07%,第5天下降至8.38%,蛭弧菌属在前3 d呈现低占比(0.08%~0.16%),第5天呈现一个峰值,占比为18.53%,第7天后逐渐下降。[结论]EPS水循环系统水处理效果良好,且推测在前5 d的水处理过程中,变形菌门和蛭弧菌属在水处理过程中发挥重要作用。
[Objective]To explore the dynamic change of microbial community in water treatment cycle.[Method]The EPS foam filter beads was used as biological filter material for water treatment test.Through the water quality index ammonia nitrogen,nitrite nitrogen,TOC to evaluate its water quality treatment effect,and using Illumina miseq high throughput of bacteria 16S rRNA gene V3+V4 region sequencing in water,to analyze the structure and diversity of microbial community structure throughout the water treatment cycle.[Result]The final removal rates of ammonia nitrogen,nitrite nitrogen and TOC could reach more than 98.0%,99.0%and 95.0%in the 10-day water treatment process,the water quality situation stabilized on the 5th day.By analyzing the changes of bacterial abundance and diversity in water at different times,it was found that the microbial richness in water was the highest on the 9th day,the community diversity was the highest on the 5th day.In the whole cycle,the dominant bacteria had little change in phylum classification,and the Proteobacteria had a major advantage in water.In the first three days,it accounted for more than 95.00%,and after the fifth day,it gradually decreased from 77.79%to 63.57%.On the level of genus classification,the bacterial richness in water was significantly different among genera.Vibrio accounted for 2.97%on the first day,rapidly increased to 98.07%on the third day,and decreased to 8.38%on the fifth day.Bdellovibrio showed a low proportion of(0.08%-0.16%)in the first three days,and a peak of 18.53%in the fifth day,and gradually decreased after the seventh day.[Conclusion]The treatment effect of EPS water circulation system is good.It is speculated that Proteobacteria and Bdellovibrio plays an important role in the water treatment in the first 5 days.
