摘要
目的:探讨清下方对急性内毒素肝损伤大鼠肝细胞凋亡的抑制作用及其作用机制。方法:采用脂多糖(Lipopolysaccharide,LPS)加D-氨基半乳糖(D-galactosamine,D-GalN)造成大鼠急性内毒素性肝损伤模型,并予以清下方进行干预,检测各组大鼠肝功能及凝血酶原时间(Prothrombin Time,PT)、肝组织病理改变,Tunel检测各组大鼠肝细胞凋亡指数,反转录聚合酶链式扩增反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)检测各组大鼠肝组织Bcl-2、Bax及Caspase-3 mRNA表达量。结果:与造模组比较,清下方可显著降低内毒素性肝损伤大鼠血谷丙转氨酶(Alanine Aminotransferase,ALT)、谷草转氨酶(Aspartate Aminotransferase,AST)、总胆红素(Total Bilirubin,TBIL)水平(P<0.05或P<0.01),改善肝组织病理结果,Tunel检测显示造模组可见较多的凋亡细胞,与对照组及清下方组相比均有显著差异(P<0.01),造模组大鼠Bax、Caspase-3 mRNA表达量显著高于对照组及清下方组(P<0.05),BCL-2 mRNA表达量则较对照组及清下方组显著降低(P<0.05)。结论:清下方可显著减轻内毒素导致的大鼠肝功能损伤,改善大鼠肝组织病理、减少肝细胞凋亡。
Objective:To investigate the effect and action mechanism of Qingxia formula(清下方)on hepatocyte apoptosis in rats with acute hepatic injury induced by lipopolysaccharide plus D-galactosamine(LPS/D-GalN).Methods:The acute hepatic injury model was established by LPS/D-GalN and then intervened with Qingxia formula.Serum liver function,PT and pathological changes of liver tissue were observed,hepatocyte apoptosis index was detected by Tunel.Bcl-2,Bax and Caspase-3 mRNA expressions of liver tissue in rats were detected by RT-PCR.Results:Compared with the model group,Qingxia formula can decrease serum ALT,AST,TBIL levels significantly(P<0.05 or P<0.01),and ameliorate pathological changes of liver tissue.The Tunel test showed that there were more apoptotic cells in the model group(P<0.01),the Bax,Caspase-3 mRNA expressions in the model group were significantly higher than those in the control group and the Qingxia formula group(P<0.05),and the Bcl-2 mRNA expression was significantly lower than that in the control group and the Qingxia formula group(P<0.05).Conclusion:Qingxia formula can significantly ameliorate liver function injury and hepatic tissue pathology,and decrease hepatocyte apoptosis in rats with endotoxin-induced liver injury.
