lncRNA SNHG1和miR-339-3p在肝细胞癌组织中的表达及临床意义

摘要 目的探究lncRNA SNHG1和miR-339-3p在肝细胞癌(HCC)组织中的表达水平及临床意义。方法收集2016年9月至2018年9月于太和医院进行手术切除治疗的108例HCC患者的肿瘤组织及癌旁组织(距肿瘤边缘>3 cm处)标本。采用实时荧光定量PCR法检测lncRNA SNHG1和miR-339-3p的表达水平,分析其与患者临床病理特征的关系,并探讨其与患者预后生存的关系。结果与癌旁组织比较,HCC组织中lncRNA SNHG1的表达水平较高,miR-339-3p的表达水平较低,2组差异均有统计学意义(P均<0.05)。lncRNA SNHG1高表达组中Edmondson分级为Ⅲ~Ⅳ级、TNM分期为Ⅲ~Ⅳ期、有乙型肝炎病史、有淋巴结转移的患者占比显著高于lncRNA SNHG1低表达组,差异均有统计学意义(P均<0.05)。miR-339-3p高表达组中Edmondson分级为Ⅲ~Ⅳ级、TNM分期为Ⅲ~Ⅳ期、有乙型肝炎病史、有淋巴结转移的患者占比显著低于miR-339-3p低表达组,差异均有统计学意义(P均<0.05)。lncRNA SNHG1高表达组的3年总生存率显著低于lncRNA SNHG1低表达组,而miR-339-3p低表达组的3年总生存率显著低于miR-339-3p高表达组,组间比较差异均有统计学意义(P均<0.05)。高Edmondson分级、高TNM分期、乙型肝炎病史、淋巴结转移、lncRNA SNHG1高表达及miR-339-3p低表达均是影响患者预后生存的独立危险因素(P均<0.05)。结论HCC组织中lncRNA SNHG1的表达水平较高,miR-339-3p的表达水平较低,且两者的表达水平与患者预后相关。 Objective An attempt is made in this paper to explore the expression and clinical significance of lncRNA SNHG1 and miR-339-3p in hepatocellular carcinoma(HCC).Methods The cancer tissues and adjacent tissues(>3 cm from the tumor edge)of 108 patients with HCC who underwent surgical resection in Taihe Hospital from September 2016 to September 2018 were collected.Real-time quantitative PCR was used to detect the expression of lncRNA SNHG1 and miR-339-3p to analyze their relationship with the clinicopathological characteristics of patients and to explore their relationship with the prognosis and survival of patients.Results Compared with adjacent tissues,the expression of lncRNA SNHG1 in HCC tissues is higher,and that of miR-339-3p in HCC tissues is lower,with statistically significant differences(P<0.05).The proportion of patients with Edmondson gradesⅢandⅣ,TNM stagesⅢandⅣ,history of hepatitis B,and lymph node metastasis in the lncRNA SNHG1 high expression group is significantly higher than that in the lncRNA SNHG1 low expression group(P<0.05).The proportion of patients with Edmondson gradesⅢandⅣ,TNM stagesⅢandⅣ,history of hepatitis B,and lymph node metastasis in the miR-339-3p high expression group is significantly lower than that in the miR-339-3p low expression group(P<0.05).The 3-year overall survival rate of patients in the lncRNA SNHG1 high expression group is significantly lower than that in the lncRNA SNHG1 low expression group,while the 3-year overall survival rate of patients in the miR-339-3p low expression group is significantly lower than that in the miR-339-3p high expression group,with statistically significant differences(P<0.05).High Edmondson classification,high TNM stage,history of hepatitis B,lymph node metastasis,high lncRNA SNHG1 expression,and low miR-339-3p expression are independent risk factors for the prognosis of patients(P<0.05).Conclusion lncRNA SNHG1 is highly expressed and miR-339-3p is low expressed in HCC,and their expressions are related to prognosis.
