摘要
目的:探讨千金子二萜醇调控TGF-β1/Smad通路影响肾癌小鼠TNF-α和IL-17表达的机制。方法:建立肾癌移植瘤小鼠,分为模型组、实验组(千金子二萜醇组)和SB-431542组(TGF-β1/Smad通路抑制剂组),实验组小鼠灌胃给予20 mg/kg千金子二萜醇溶液,模型组灌胃给予等体积生理盐水,SB-431542组腹腔注射10 mg/kg SB-431542溶液,计算脾脏指数;Western blot检测Smad2/3和p-Smad2/3蛋白表达;ELISA检测肾癌移植瘤小鼠血清TNF-α和IL-17表达。结果:给药19 d后,与模型组相比,实验组和SB-431542组小鼠脾脏指数降低(P<0.05),Smad2/3蛋白表达升高(P<0.05),p-Smad2/3蛋白表达、TNF-α和IL-17水平明显降低(P<0.01)。结论:千金子二萜醇可通过调控TGF-β1/Smad通路活性降低肾癌小鼠TNF-α和IL-17表达。
Objective:To explore mechanism of lathyrol regulating expressions of TNF-α and IL-17 through TGF-β1/Smad pathway in renal cell carcinoma mice.Methods:Transplanted tumor mice were constructed and divided into model group,experimental group(Lathyrol group)and SB-431542 group(TGF-β1/Smad pathway inhibitor group). Mice in experimental group were given20 mg/kg Lathyrol solution,model group was given equal volume of normal saline,and SB-431542 group was intraperitoneally injected with 10 mg/kg SB-431542 solution to calculate spleen index. Expressions of Smad2/3 and p-Smad2/3 proteins were detected by Western blot;expressions of TNF-α and IL-17 in serum of transplanted renal cell carcinoma mice were detected by ELISA.Results:After 19 d of administration,compared with model group,spleen index were decreased in experimental group and SB-431542 group(P<0.05),expressions of Smad2/3 protein were increased(P<0.05),expressions of p-Smad2/3 protein,TNF-α and IL-17 levels were decreased significantly(P<0.01).Conclusion:Lathyrol can reduce expressions of TNF-α and IL-17 in renal cell carcinoma mice through regulating activity of TGF-β1/Smad pathway.
