摘要
为了研究皮下埋植褪黑素(MT)对奶牛超数排卵的影响,本试验选择48头发育良好的荷斯坦牛分为对照组(0mg MT)和试验组(160mg MT、240mg MT),结合CIDR-FSH-PG-GnRH的方案进行超数排卵。通过B超检测卵巢及非手术法冲胚效果,统计超数排卵期间卵巢长、宽、高及回收胚胎情况与胚胎移植妊娠率。结果表明,皮下埋植160mg MT的效果较好,输精时卵泡数显著高于0mg、240mg组(P<0.05),卵巢宽度及高度显著大于0mg、240mg组(P<0.05),冲胚时黄体数显著高于0mg、240mg组(P<0.05),卵泡数、卵巢长度及高度呈增大的趋势,但差异不显著(P>0.05)。在胚胎生产及移植方面,显著提高了可用胚胎数及比例、A级胚数及比例、移植妊娠率(P<0.05),显著降低了未受精胚数及比例、退化胚数及比例、B级胚数及比例(P<0.05),冲胚总数、囊胚数、早期囊胚数、桑葚胚数均有上升的趋势(P>0.05)。综上,皮下埋植160mg褪黑素能够提高可用胚胎数、胚胎质量和胚胎移植妊娠率,从而提高供体奶牛超数排卵效果。
To study the effect of subcutaneous implantation of melatonin(MT)on superovulation in dairy cows.In this experiment,48 well-developed Holstein dairy cows were selected for subcutaneous implantation of melatonin experiment.The experiment was divided into control group(0mg MT)and experimental group(160mg MT,240mg MT)combined with CIDR-FSH-PG-GNRH regimen for superovulation.Ovary were detected by B-ultrasound and embryo recovery was performed by non-surgical method.The length,width and height of ovary,embryo recovery and embryo transfer pregnancy rate during superovulation were counted.The results showed that in terms of ovarian response implantation of 160 mg MT has significantly differences in the number of follicles(P<0.05),the width(P<0.05)and height(P<0.05)of ovary during first artificial insemination(9.8d),but no significant differences in ovarian length(P>0.05).The luteal number(P<0.05)and the width(P<0.05)were significantly different during embryo recovery(16d).The number of follicles(P>0.05),the length(P>0.05)and the height(P>0.05)of ovary were increased,but the differences were not significant during embryo recovery(16d).In embryo production and transplantation,the number and proportion of available embryos,the number and proportion of A-grade embryos,and the pregnancy rate of transplantation were significantly increased(P<0.05),while the number and proportion of unfertilized embryos,the number and proportion of degenerated embryos,and the number and proportion of B-grade embryos were significantly decreased(P<0.05).The total number of embryo recovery,blastocyst number,early blastocyst number and morula number were rising trend(P>0.05).In conclusion,subcutaneous implantation of 160 mg melatonin can improve the number of available embryos,embryo quality and embryo transfer pregnancy rate,therefore improving the results of superovulation of dairy cows.
