摘要
目的:探讨针刺是否通过调节脑组织ICAM-5 mRNA的表达从而发挥脑保护作用。方法:采用雄性SD大鼠构建大脑中动脉闭塞再灌注损伤(MCAO)模型,将大鼠随机分为4组,分别为正常组(A组)、假手术组(B组)、MCAO模型组(C组)与MCAO模型+针刺组(D组)。采用实时荧光定量PCR(RT-qPCR)检测缺血区脑组织ICAM-5 mRNA表达,并根据HE染色和TUNEL检测神经元凋亡水平以评估治疗效果。结果:针刺可改善缺血性脑卒中大鼠神经细胞形态及减少神经元凋亡;RT-qPCR结果显示:C组、D组脑组织ICAM-5 mRNA表达水平较A组和B组均下降,而与C组比较,D组脑组织ICAM-5 mRNA表达明显上调(P<0.01)。A组和B组比较差异无统计学意义(P>0.05)。结论:针刺治疗缺血性中风的有效性可能是通过上调ICAM-5 mRNA表达水平从而达到神经保护的作用。
Objective:To investigate whether acupuncture can protect brain by regulating the expression of ICAM-5 mRNA in brain tissue.Methods:The model of middle cerebral artery occlusion(MCAO)reperfusion injury was established in SD male rats.The rats included in the experiment were randomly divided into the normal group(group A),the sham operation group(group B),the model group(group C)and the acupuncture group(group D).Real-time fluorescent quantitative PCR(RT-qPCR)was used to detect the expression of ICAM-5 mRNA in brain tissue of the ischemic area,and HE staining and TUNEL were used to detect the level of neuronal apoptosis to evaluate therapeutic effect.Results:Acupuncture could improve neuronal morphology and reduce neuronal apoptosis in rats with ischemic stroke.RT-qPCR results showed that compared with that in group A and group B,the expression of ICAM-5 mRNA was decreased in group C and group D;compared with that in group C,the expression of ICAM-5 mRNA was significantly up-regulated in group D(P<0.05);in which there was no statistical difference between group A and group B(P>0.05).Conclusion:The effectiveness of acupuncture in treatment of ischemic stroke may achieve neuroprotection by up-regulating the expression of ICAM-5 mRNA.
