不同煮散粒度下加味大柴胡汤中各成分影响及其治疗糖尿病作用机制研究 被引量:3

Study on the Effect of Various Components in Jiawei Dachaihu Decoction(加味大柴胡汤)Under Different Boiled Powder Sizes and Its Mechanism in Treating Diabetes

摘要 目的以加味大柴胡汤为研究对象,网络药理学为基础,研究不同煮散粒度对其水煎液中各种成分含量的影响及其治疗糖尿病的作用机制。方法采用蒽酮-硫酸显色法、高效液相色谱法,以总多糖含量及芒果苷、柚皮苷、盐酸巴马汀、黄芩苷、汉黄芩苷为指标性成分,考察不同煮散粒度下加味大柴胡汤水煎液的化学成分;用CTD数据寻找加味大柴胡汤中5种化学成分的靶标基因构建“化学成分-靶标”图;通过GeneCards、OMIM、Drug Bank疾病数据库搜索糖尿病基因,将化学成分与疾病的交集靶标导入String数据库,其结果用Cytoscape软件进一步筛选得核心靶标和拓扑分析得蛋白相互作用(PPI)图;运用DAVID数据库对核心靶标进行通路富集分析。结果(1)经分析不同粒度下加味大柴胡汤煎煮后化学成分存在差异。(2)不同粒度下总多糖含量不同,从高到低依次为粗粉、细粉、饮片及最细粉。其中粗粉、细粉、最细粉的总多糖含量约是饮片的1.2、1.15、0.93倍。(3)与正常饮片相比,粗粉与细粉中5种化学成分均有所增加,而最细粉中5种化学成分既有增加也有下降,其中粗粉中芒果苷、柚皮苷、盐酸巴马汀、黄芩苷、汉黄芩苷分别是饮片的1.19、4.02、1.57、2.38、1.83倍;细粉中分别是饮片的1.12、3.46、1.37、2.02、1.56倍,最细粉中分别是饮片的0.11、1.94、0.65、1.06、1.00倍。(4)加味大柴胡汤中5种化学成分对糖尿病治疗具有很好的疗效,主要通过作用于PIK3CA、MAPK3、MAPK1、AKT1、PIK3R1等靶标以及癌症、乙型肝炎、癌症蛋白多糖、PI3K-Akt等信号通路达到治疗糖尿病作用。结论(1)不同粒度下加味大柴胡汤煎煮后化学成分存在差异。(2)适宜的煮散粒度其化学成分远远高于普通饮片,经分析加味大柴胡汤中各药物粒度在2.00~4.75 mm条件下各化学成分含量最高。(3)加味大柴胡汤中的黄芩苷、盐酸巴马汀等5种成分可以通过多靶标(PIK3CA、MAPK3、MAPK1、AKT1、PIK3R1等)、多通路(癌症、乙型肝炎、癌症蛋白多糖、PI3K-Akt等信号通路)起到治疗糖尿病的作用。 Objective Taking Jiawei Dachaihu Decoction(加味大柴胡汤)as the research object,based on network pharmacology,the effect of different boiled powder particle size on the content of various ingredients in its decoction and its mechanism of action on diabetes were studied.Methods Using anthrone-sulfuric acid colorimetric method and HPLC method,the chemical constituents of the Jiawei Dachaihu Decoction with different boiled powder sizes were investigated with total polysaccharide content,mangiferin,naringin,palmatine hydrochloride,baicalin and hanbaicalin as indicators.CTD data were used to find the target genes of 5 chemical components in Jiawei Dachaihu Decoction to construct the“chemical components-target”map.The diabetes genes were searched by GeneCards,OMIM and Drug Bank disease data,and the intersection targets of chemical components and diseases were imported into String database.The results were further screened by Cytoscape software to get the core target and topological analysis to get the PPI map.DAVID database was used for pathway enrichment analysis of core targets.Results(1)After analysis,there were differences in chemical composition of Jiawei Dachaihu Decoction with different particle sizes.(2)The contents of total polysaccharides were different under different particle sizes.From high to low,the total polysaccharides were coarse powder,fine powder,decoction pieces and the finest powder.The total polysaccharide content of coarse powder,fine powder and the finest powder was about 1.2,1.15 and 0.93 times of that of decoction pieces.(3)Compared with normal decoction pieces,the five chemical constituents in coarse powder and fine powder increased,while the five chemical constituents in the finest powder both increased and decreased.The mangiferin,naringin,palmatine hydrochloride,baicalin and hanbaicalin in crude powder were 1.19,4.02,1.57,2.38 and 1.83 times of that in decoction pieces,respectively.The fine powder was 1.12,3.46,1.37,2.02,1.56 times of the yinpian,and the finest powder was 0.11,1.94,0.65,1.06,1.00 times of the yinpian,respectively.(4)The five chemical components in the jiawei dachaihu decoction have a good effect on the treatment of diabetes,mainly by acting on PIK3CA,MAPK3,MAPK1,AKT1,PIK3R1and other targets,as well as cancer,hepatitis B,proteoglycan in cancer,PI3K-Akt and other signaling pathways.Conclusion(1)There are differences in chemical composition of Jiawei Dachaihu Decoction with different particle size.(2)The chemical composition of the suitable boiling powder particle size was much higher than that of ordinary decoction pieces,and the chemical composition content was the highest under the particle size of 2.00-4.75 mm in Jiawei Dachaihu Decoction.(3)Baicalin,palmatine hydrochloride and other 5 components in jiawei dachaihu decoction can play a role in the treatment of diabetes through multi-target(PIK3CA,MAPK3,MAPK1,AKT1,PIK3R1,etc.),multi-pathway(cancer,hepatitis B,proteoglycan in cancer,PI3K-Akt and other signaling pathways).
