摘要
建立了一种聚乙二醇/无机盐双水相萃取-高效液相色谱波长切换法测定饮料中7种合成着色剂的方法。样品调节pH后,加入聚乙二醇(PEG)2000和硫酸铵,经超声溶解,涡旋提取,离心后,取上层溶液进行高效液相色谱分析。色谱柱采用C柱,流动相为0.02mol/L乙酸铵水溶液-甲醇,梯度洗脱,检测波长采用时间程序波长切换法,各物质均采用各自的特征吸收波长测定。结果显示,7种合成着色剂在0.5~50μg/mL范围内线性关系良好,相关系数均大于0.9999,方法检出限为2.76~12.7μg/kg,定量限为9.20~42.2μg/kg,平均加标回收率为83.6%~98.9%,相对标准偏差(RSD)为1.27%~3.32%。本方法前处理简便快速,回收率高,重复性好,适合于饮料中7种合成着色剂的测定。
A polyethylene glycol/inorganic salt aqueous two-phase extraction with high performance liquid chromatography wavelength switching method was established for the determination of 7 synthetic colorants in beverages. After adjusting the pH,polyethylene glycol(PEG)2000 and ammonium sulfate were added and dissolved by ultrasound,then after vortex and centrifuge,the upper layer solution was taken for HPLC analysis.The HPLC conditions were :Cchromatographic column,mobile phase 0.02mol/L ammonium acetate aqueous solution-methanol,and gradient elution. The detection wavelength adopted wavelength switching time program,and each substance was measured with its own characteristic absorption wavelength. The results showed that seven synthetic colorants had a good linear relationship in the range of 0.5 ~ 50μg/mL;the correlation coefficients were all greater than 0.9999,the detection limit of the method was 2.76 ~ 12.7μg/kg,and the limit of quantification was 9.20 ~ 42.2μg/kg. The standard recovery was 83.6% ~ 98.9%,and the relative standard deviation(RSD)was 1.27% ~ 3.32%. This method is simple and rapid,has high recovery and good repeatability. It is suitable for the determination of seven synthetic colorants in beverages.
