ELOVL2和MCP-1基因多态性与肺结核易感性的关联 被引量:3

Association of ELOVL2 and MCP-1 gene polymorphisms with susceptibility to pulmonary tuberculosis

摘要 目的分析研究ELOVL脂肪酸延长酶基因2(ELOVL2)、单核细胞趋化因子蛋白(MCP-1)基因多态性与肺结核易感性的关联性。方法选择2017年12月-2020年12月三台县人民医院收治的103例肺结核患者作为肺结核组,同时选取同期健康体检者103名作为对照组,提取两组受试者外周血DNA,采用聚合酶链式反应(PCR)产物直接测序法检测ELOVL2、MCP-1基因位点单核苷酸多态性(SNPs),运用Logistic回归分析ELOVL2、MCP-1基因多态性与肺结核易感性的关联性。结果两组ELOVL2基因rs3798719位点CC、CT和TT基因型及C、T等位基因频率差异有统计学意义(P<0.05);MCP-1基因-2518位点AA、GA、GG基因型及A、G等位基因频率差异有统计学意义(P<0.05);ELOVL2基因rs3798719位点超显性、隐性模型在肺结核组和对照组之间,差异有统计学意义(P<0.05);两组MCP-1基因-2518位点超显性、显性及隐性模式差异有统计学意义(P<0.05);多因素Logistic回归分析结果显示,ELOVL2基因rs3798719位点、MCP-1基因-2518位点多态性是肺结核易感性的影响因素(P<0.05)。结论ELOVL2基因rs3798719位点、MCP-1基因-2518位点多态性与肺结核易感性有关。 OBJECTIVE To analyze the relationship between gene polymorphisms of ELOVL fatty acid elongase 2(ELOVL2)and monocyte chemoattractant protein-1(MCP-1)and susceptibility to pulmonary tuberculosis.METHODS Totally 103 patients with pulmonary tuberculosis admitted to the hospital between Dec.2017 and Dec.2019 were selected as PTB group,and another 103 healthy people with physical examination during the same period were included in control group.The peripheral blood DNA of the two groups of subjects was extracted and polymerase chain reaction(PCR)product direct sequencing method was used to detect the single nucleotide polymorphisms(SNPs)at loci of ELOVL2 and MCP-1 genes.The relationship between gene polymorphisms of ELOVL2 and MCP-1 and susceptibility to pulmonary tuberculosis was analyzed by Logistic regression analysis.RESULTS The CC,CT and TT genotypes and the frequency of C and T alleles at rs3798719 of ELOVL2 gene were significantly different between the two groups(P<0.05).The AA,GA,GG genotypes and the frequencies of A and G alleles at 2518 of McP-1 gene were significantly different(P<0.05).There were statistically significant differences between the PTB group and the control group in the dominant and recessive models of RS3798719 locus of ELOVL2 gene(P<0.05).There were statistically significant differences in the superdominant,dominant and recessive patterns of McP-1 gene-2518 locus between the two groups(P<0.05).Multivariate Logistic regression analysis showed that polymorphism of RS3798719 locus of ELOVL2 gene and McP-1 locus 2518 were the influencing factors of pulmonary tuberculosis susceptibility(P<0.05).CONCLUSION The polymorphisms at rs3798719 locus of ELOVL2 gene and at-2518 locus of MCP-1 gene are related to the susceptibility to pulmonary tubercuPTBlosis.
