杜仲补天素丸对环磷酰胺诱导的生精障碍保护作用 被引量:1

摘要 目的研究不同剂量杜仲补天素丸对腹腔注射环磷酰胺致小鼠生精障碍的改善作用。方法60只雄性小鼠随机分为空白对照组、模型组、阳性药生精胶囊组(0.8 g/kg)、杜仲补天素丸高剂量组(2.2 g/kg)、中剂量组(1.1 g/kg)、低剂量组(0.55 g/kg)。采用腹腔注射环磷酰胺CP(60 mg/kg)连续7d,制备生精障碍小鼠模型。造模后连续灌胃给药4周,每周检测小鼠体重。灌胃结束后,摘眼球取血并立即取出脏器待测,并对精子质量进行检测。ELISA法测定血清中促卵泡素(FSH)、睾酮(T)、促黄体生成素(LH),血清超氧化物歧化酶(SOD)、丙二醛(MDA),还原型谷胱甘肽(GSH)水平。结果与模型组比较,杜仲补天素丸高、中、低剂量组能升高小鼠体重,升高生殖器官指数,增加精子总数,提高精子活率,降低精子畸形率,改善血清中促卵泡素(FSH)、睾酮(T)、促黄体生成素(LH)水平,升高血清超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)水平,降低丙二醛(MDA)水平。结论杜仲补天素丸能够改善环磷酰胺造成的生精障碍,其作用机制可能与抗氧化作用有关。 Objective To study the effect of different doses of Duzhongbutiansuwan on improving spermatogenesis in mice induced by intraperitoneal injection of cyclophosphamide. Methods 60 male mice were randomly divided into a blank control group, a model group, a positive medicine Shengjing capsule group(0.8 g/kg), Duzhongbutiansuwan high-dose group(2.2 g/kg), middle-dose group(1.1 g)./kg), and low-dose group(0.55 g/kg). Cyclophosphamide CP(60 mg/kg) was injected intraperitoneally for 7 days to prepare a mouse model of spermatogenesis disorder. After making the model, intragastric administration was continued for 4 weeks, and the weight of the mice was measured every week. After the gavage, the eyeballs were removed to take blood and the organs were immediately taken out for testing, and the sperm quality was tested. ELISA method to determine serum levels of follicle stimulating hormone(FSH), estosterone(T), luteinizing hormone(LH), serum superoxide dismutase(SOD), malondialdehyde(MDA), and reduced glutathione(GSH). Results Compared with the model group, the high, medium, and low dose groups of Duzhongbutiansuwan can increase the weight of mice, increase the index of reproductive organs, increase the total number of sperm, increase sperm motility, reduce the rate of sperm abnormalities, and improve serum follicle stimulating hormone(FSH), testosterone(T), luteinizing hormone(LH) levels, increase serum superoxide dismutase(SOD) and reduced glutathione(GSH) levels, and reduce malondialdehyde(MDA) levels. Conclusion Duzhongbutiansuwan can improve spermatogenesis caused by cyclophosphamide, and its mechanism of action may be related to its antioxidant effect.
