初诊弥漫大B细胞淋巴瘤患者骨髓来源抑制性细胞的亚群分布及功能异常分析 被引量:1

摘要 目的 探讨初诊的弥漫大B细胞淋巴瘤(DLBCL)患者外周血和骨髓中骨髓来源抑制性细胞(MDSC)亚群的分布和功能。方法 收集50例初诊DLBCL患者和30例健康供者的外周血标本,10例初诊DLBCL患者和10例健康供者的骨髓标本。采用多色流式细胞术检测上述标本中MDSC两大亚群即单核细胞性MDSC(M-MDSC)和粒细胞性MDSC(G-MDSC)的分布情况。从收集的初诊DLBCL患者和健康供者外周血标本中分别随机选取20例,通过DCFH-DA探针法检测其外周血标本M-MDSC和G-MDSC的活性氧(ROS)水平。结果 初诊DLBCL患者外周血M-MDSC比例明显高于健康供者,差别有统计学意义(P=0.001);2组间G-MDSC比较,差别无统计学意义(P=0.770)。初诊DLBCL患者骨髓标本中M-MDSC比例亦高于健康供者,差别有统计学意义(P=0.001);2组间G-MDSC比例比较,差别无统计学意义(P=0.213)。G-MDSC的ROS表达水平明显高于健康供者,差别有统计学意义(P=0.005);2组间M-MDSC的ROS表达水平比较,差别无统计学意义(P=0.099)。结论 初诊DLBCL患者的外周血、骨髓中M-MDSC亚群比例的上调和外周血G-MDSC的ROS表达水平增高可能参与构成异常免疫微环境,介导疾病进展过程。 Objective To investigate the distribution and function of myeloid-derived suppressor cell(MDSC)subsets in peripheral blood and bone marrow of newly diagnosed diffuse large B-cell lymphoma(DLBCL)patients.Methods Peripheral blood samples were collected from 50 newly diagnosed DLBCL patients and 30 healthy donors.Bone marrow samples were collected from 10 newly diagnosed DLBCL patients and 10 healthy donors.The distribution of two subsets of MDSC,mononuclear MDSC(M-MDSC)and granulocytic MDSC(G-MDSC),was detected by multi-color flow cytometry.M-MDSC and G-MDSC reactive oxygen species(ROS)levels in 20 newly diagnosed DLBCL patients and 20 healthy donors were detected by the DCFH-DA probe method.Results The percentage of M-MDSC in the peripheral blood of newly diagnosed DLBCL patients was significantly higher than that of healthy donors(P=0.001).There was no significant difference in G-MDSC between the two groups(P=0.770).The proportion of M-MDSC in bone marrow samples of newly diagnosed DLBCL patients was also higher than that of healthy donors,and the difference was statistically significant(P=0.001).The percentage of G-MDSC between the two groups had no significant difference(P=0.213).In addition,the ROS level of G-MDSC was significantly higher than that of healthy donors(P=0.005).However,there was no significant difference in the ROS level of M-MDSC between the two groups(P=0.099).Conclusion The up-regulation of M-MDSC subsets in peripheral blood and bone marrow and the increase of G-MDSC expression ROS level in peripheral blood of newly diagnosed DLBCL patients may participate in the formation of the abnormal immune microenvironment and mediate the progression of the disease.
