组蛋白脱乙酰基酶9对结直肠癌增殖和转移能力的影响和机制研究

Effects of histone deacetylase 9 on proliferation and metastasis of colorectal cancer and the mechanism

摘要 目的探讨组蛋白脱乙酰基酶9(HDAC9)对结直肠癌(CRC)细胞增殖和转移的影响,探讨其发挥作用的可能机制。方法选取2017年1月至2017年6月间西安医学院第二附属医院行手术切除,并经病理确认的新鲜CRC癌组织和其配对的非癌组织6例。采用qPCR、western blotting检测HDAC9在CRC组织和细胞系SW480、SW620和HCT116中的表达水平,选择高表达的CRC细胞系进行HDAC9 siRNA感染,分为si-NC组、si-HDAC9-1组si-HDAC9-2组。qPCR、western blotting检测各组细胞中HDAC9的表达水平;MTS和平板克隆实验检测HDAC9对细胞增殖的影响;Transwell实验检测HDAC9对细胞转移能力的影响;western blotting检测HDAC9对PI3K/AKT信号通路的影响。结果qPCR、western blotting结果显示,HDAC9在CRC组织和细胞系的表达上调,差异有统计学意义(P<0.05)。选择表达最高的CRC细胞系SW480转染HDAC9 siRNA,western blotting结果显示,si-HDAC9-1、si-HDAC9-2组细胞中HDAC9的表达降低,差异有统计学意义(P<0.05)。与si-NC组相比,si-HDAC9-1和si-HDAC9-2组SW480细胞增殖和转移能力下降,且细胞中pPI3K和pAKT蛋白的表达降低。结论干扰HDAC9的表达抑制CRC细胞增殖和转移能力。HDAC9可能是治疗CRC的新型靶标。 Objective To investigate the effects of histone deacetylase 9(HDAC9)on proliferation and metastasis of colorectal cancer(CRC)cells,and to explore the possible mechanisms of its function.Methods From January 2017 to June 2017,fresh pathology confirmed CRC tissues and the matched tissues were collected from 6 patients undergoing resection at The Second Affiliated Hospital of Xi’an Medical College.qPCR and western blotting were used to detect the expression of HDAC9 in CRC tissues and cell lines SW480,SW620 and HCT116.HDAC9 highly expressed CRC cell lines were selected for HDAC9 siRNA infection,which were divided into si-NC group and si-HDAC9-1 group,si-HDAC9-2 group.qPCR and western blotting were used to detect the expression of HDAC9 in each group.MTS and plate cloning assays were used to detect the effect of HDAC9 on cell proliferation.Transwell assay was used to detect the effect of HDAC9 on cell metastasis.Western blotting was used to detect the effect of HDAC9 on PI3 K/AKT signaling pathway.Results qPCR and western blotting showed that the expression of HDAC9 was up-regulated in CRC tissues and cell lines(all P<0.05).HDAC9 siRNA was transfected into the highest expression of CRC cell line SW480.The results of qPCR and western blotting showed that the expression of HDAC9 were decreased in si-HDAC9-1 and si-HDAC9-2 groups(all P<0.05).Compared with si-NC group,the proliferation and metastasis ability of SW480 cells were decreased in both si-HDAC9-1 and si-HDAC9-2 groups,and the expression of pPI3 K and pAKT proteins were reduced in si-HDAC9-1 and si-HDAC9-2 groups cells.Conclusion Interfering with the expression of HDAC9 inhibits the proliferation and metastasis of CRC cells.HDAC9 may be a novel target for the treatment of CRC.
