大黄提取物激活p38MAPK促进HepG2肝癌细胞凋亡 被引量:3

摘要 目的探讨中药大黄提取物对HepG2肝癌细胞增殖与凋亡的影响和作用机制。方法采用MTS、平板克隆形成实验、钙黄绿素/PI染色观察大黄提取物对HepG2肝癌细胞活性的影响,然后通过Hoechst33342/PI染色观察大黄提取物对肝癌细胞凋亡的影响,最后采用蛋白印迹检测大黄提取物对肝癌细胞内p38MAPK以及凋亡相关蛋白Bcl-2、Bax表达水平的影响。结果MTS、平板克隆形成实验、钙黄绿素/PI染色提示大黄提取物显著抑制肝癌细胞的活性。Hoechst33342/PI染色提示大黄提取物能够浓度依赖性地促进肝癌细胞凋亡。此外,大黄提取物处理后显著增加肝癌细胞内p38MAPK的磷酸化水平,上调凋亡相关蛋白Bax的表达水平并下调Bcl-2表达。使用p38MAPK抑制剂能部分减弱大黄提取物的抗肝癌作用。结论大黄提取物具有抑制肝癌细胞活性的作用,其作用机制可能是通过激活p38MAPK信号通路诱导肝癌细胞凋亡发挥的。 Objective To explore the anti-tumor effect and mechanism of rhubarb extract on human hepatocellular carcinoma cells.Methods The cell proliferation ability were detected by MTS and colony formation assays.Live/dead assay was detected by Calcein/PI staining and cell apoptosis was detected by Hoechst33342/PI staining.P38MAPK and apoptosis proteins in hepatocellular carcinoma cells in response to RP were determined by Western blot.Results MTS,colony formation assays and Calcein/PI staining indicated that rhubarb extract significantly inhibited the proliferation of HepG2.Hoechst33342/PI staining suggested that rhubarb extract could induce apoptosis of HepG2 in a concentration-dependent manner.In addition,treatment with rhubarb extract significantly increased the phosphorylation level of p38MAPK,up-regulated the expression level of apoptosis-related protein Bax and down-regulated the expression of Bcl-2.Moreover,inhibition of p38MAPK partly abrogates the inhibitory effects of RP in HepG2 cells.Conclusion Our results indicate that rhubarb extract can inhibit the proliferation and induce apoptosis of hepatocellular carcinoma cells,and its mechanism of action may be exerted by activating the p38MAPK signaling pathway.
