摘要
肿瘤是发病机制复杂、预后较差、发病率和病死率较高的一类疾病。目前临床上多采用手术、化疗、放疗等手段进行治疗。中医药是防治恶性肿瘤的有效手段之一,在产生抗肿瘤效应的同时,还能够有效改善患者的临床症状,减轻放化疗毒副作用,逆转耐药,提高生活质量等。网络药理学作为一门新兴学科,是在系统生物学的基础上,通过构建“药物-靶点-疾病”网络,阐述药物作用于疾病的靶点和通路,与中药防治恶性肿瘤的多成分、多靶点的效应不谋而合,已广泛用于抗肿瘤中药研究;研究显示抗肿瘤中药在抑制肿瘤细胞增殖、诱导细胞自噬和凋亡、抑制肿瘤细胞侵袭和迁移、调节机体免疫功能、抑制肿瘤血管生成以及调节肿瘤微环境等方面起到了重要的作用。因此,本文对网络药理学在防治肿瘤疾病的相关研究进行归纳和总结,以期为中药抗肿瘤的研究提供新的思路和参考。
Tumor is a disease with complex pathogenesis, poor prognosis, high morbidity and mortality. At present, surgery, chemotherapy, radiotherapy and other methods are clinically applied in treatment of tumor. TCM is one of the effective means for prevention and treatment of malignant tumor. It can not only produce anti-tumor effects, but also effectively improve clinical symptoms, reduce adverse reactions of chemotherapy and radiotherapy, reverse drug resistance and improve life quality of the patients. Network pharmacology, as an emerging discipline, illustrates the target and signaling pathway of medications based on systematic biology and the construction of ‘drug-target-disease’ network. It coincides with the multi-component and multi-target of TCM in preventing and treating malignant tumor, and has been widely used for TCM studies on anti-tumor. Studies have shown that Chinese medicinal, which have anti-tumor effects, play an important role in inhibiting cell proliferation, inducing autophagy and apoptosis, inhibiting cell invasion and migration, regulating immune function, inhibiting tumor angiogenesis and regulating tumor micro-environment. Therefore, this study summarized the related studies of network pharmacology in prevention and treatment of tumor-related diseases, in order to provide new clues and reference for the study of Chinese medicinal in anti-tumor.
