摘要
针对藤仓赤霉菌的生长及其合成赤霉素GA 3的代谢特征,添加不同的表面活性剂和维生素,考察其对藤仓赤霉菌的生长和赤霉素合成的影响,获得最适的表面活性剂和维生素添加种类,发现吐温-80、维生素B_(6)、维生素B_(12)、生物素有利于GA_(3)的合成,并对其添加量进行了优化。进一步建立了一种残糖反馈补料发酵工艺,发现葡萄糖浓度对GA_(3)的合成影响较大,发酵过程中控制残留葡萄糖浓度为30~40 g/L时最有利于GA_(3)的合成。在此操作条件下,控制初糖浓度为40 g/L,进行了补料发酵,最终GA_(3)的发酵产量达到了575.13 mg/L,与分批发酵相比提高了13.86%。
According to the growth of Fusarium fujikuroi and its metabolic characteristics for gibberellin GA_(3)biosynthesis,different surfactants and vitamins were added to investigate their effects on the growth of Fusarium fujikuroi and synthesis of gibberellin,and the optimum types of surfactants and vitamins were obtained.It was found that Tween-80,vitamin B 6,vitamin B_(12)and biotin were beneficial to the synthesis of GA_(3),and their addition amounts were optimized.Further,a residual glucose feedback fed-batch fermentation process was established,and it was found that the glucose concentration had a great effect on GA_(3)biosynthesis,and the control of the residual glucose concentration to 30-40 g/L during the fermentation process was most beneficial to the synthesis of GA_(3).Under this operating conditions,the initial sugar concentration was controlled at 40 g/L,and the fed-batch fermentation was performed,the final fermentation yield of GA_(3)reached 575.13 mg/L,which increased by 13.86%compared with the batch fermentation.
