摘要
以不同贮藏期的‘空心李’果实为试材,利用超高效液相色谱串联质谱(UPLC-MS/MS)广泛靶向代谢组学方法鉴定果实中初生代谢物,并通过样本相关性分析、主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)等方法筛选出不同贮藏期的差异代谢物。结果表明:不同贮藏期的‘空心李’果实共检测出6类307种初生代谢物,其中存在显著差异代谢物114种,分别是27种氨基酸及其衍生物、17种核苷酸及其衍生物、34种脂质、21种有机酸、10种糖及醇类代谢物和5种维生素。各组间的差异代谢物注释到57条通路,有9条显著富集的代谢通路,分别为嘌呤代谢、ABC转运器、丙酸代谢、硫代葡萄糖苷生物合成、氨基酸类代谢通路,这些代谢通路可能是‘空心李’果实贮藏过程中品质变化的关键代谢通路。该研究结果可为‘空心李’果实品质检测和评价提供理论依据和技术支撑。
The primary metabolites in‘Kongxin’plum(Prunus salicina)fruits of different storage periods were identified and compared using UPLC-MS/MS widely targeted metabolomics method.The differential metabolites during storage were screened by sample correlation analysis,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA).The results illustrated that a total of 307 primary metabolites were detected in six categories,of which 114 metabolites were significantly differential,including 27 amino acids and their derivatives,17 nucleotides and their derivatives,34lipids,21 organic acids,10 sugars and alcohols,and 5 vitamins,in the‘Kongxin’plum fruits during storage.The differential metabolites between each group were annotated to 57 metabolic pathways,and 9 significantly differential metabolic pathways were identified,of which five were identified as amino acid metabolic pathways,including purine metabolism,ABC transporters,propanoate metabolism,glucosinolate biosynthesis,amino acid metabolic pathways.These metabolic pathways may be the key metabolic pathways for the quality change of‘Kongxin’plum during storage.The results can provide theoretical basis and technical support for fruit quality detection and evaluation of‘Kongxin’plum.
