摘要
目的:验证环境监测用胰酪大豆胨琼脂培养基(Tryptone Soya Agar,TSA)中和消毒剂的效能。方法:配制7种常见环境微生物菌悬液,每一种菌的理论配制浓度为200 CFU/mL,对配制后的菌悬液进行平皿菌落计数。模拟生产车间人员消毒行为,在距离TSA平皿水平距离大于100 cm、垂直距离40 cm的位置,喷洒消毒剂(75%乙醇和70%无菌异丙醇)进行手部消毒,分别在消毒前后用菌悬液涂布TSA平皿,同时分别设置阴性对照组和空白对照组,用独立样本t检验方法对培养基菌落进行统计学分析。结果:金黄色葡萄球菌、铜绿假单胞菌、枯草芽孢杆菌、白色念珠菌、黑曲霉、表皮葡萄球菌、婴儿芽孢杆菌的实际浓度分别为270 CFU/mL、240 CFU/mL、220 CFU/mL、220 CFU/mL、150 CFU/mL、300 CFU/mL、330 CFU/mL。通过统计学分析,各实验处理组的菌落计数结果与空白对照组差异不显著(P>0.05)。结论:当前公司车间人员的日常手部消毒方式对环境监测用胰酪大豆胨琼脂培养基造成的消毒剂残留不影响环境微生物的培养,环境监测结果真实可靠。
Objective:To validate the efficacy of neutralizing disinfectant with Tryptone Soya Agar(TSA)for environmental monitoring.Methods:Seven common environmental microbial suspensions are prepared,the theoretical formulation concentration of each bacterium is 200 CFU/mL,and the configured bacterial suspension is plated with colony counting.To simulate the disinfection behavior of production workshop personnel,spray disinfectant(75%ethanol and 70%sterile isopropanol)for hand disinfection at a horizontal distance greater than 100 cm and a vertical distance of 40 cm from the TSA plate.Coat the TSA flat dish with bacterial suspension before and after disinfection,and set up a negative control group and a blank control group respectively,and statistically analyze the colonies of the medium by independent sample t-test method.Results:The colony numbers of Staphylococcus aureus,Pseudomonas aeruginosa,Bacillus subtilis,Candida albicans,Aspergillus niger,Staphylococcus epidermidis and Bacillus infantile are 270 CFU/mL,240 CFU/mL,220 CFU/mL,220 CFU/mL,150 CFU/mL,300 CFU/mL and 330 CFU/mL,respectively.Through statistical analysis,the colony counting results of each experimental treatment group are not significantly different from those in the blank control group(P>0.05).Conclusion:The daily hand disinfection method of the company’s workshop personnel does not affect the cultivation of environmental microorganisms caused by the disinfectant residue caused by pancreatic cheese soybean peptone agar medium for environmental monitoring,and the environmental monitoring results are true and reliable.
