猪丹毒丝菌C43-5株菌种毒力检定标准再评价

摘要 为进一步完善猪丹毒疫苗毒力检定标准,对猪丹毒丝菌C43-5株在肉肝胃消化汤中的生长规律进行了分析,测定了菌株对猪的最小致死剂量,并对1986、1996、2006、2015年4个冻干代次菌种进行了毒力和免疫原性测定。结果显示:C43-5株在肉肝胃消化汤中培养16 h时活菌数最高,而后进入稳定期,培养19 h后活菌数迅速下降,菌体大量死亡;以3.5×10^(10) CFU活菌静脉注射56~63日龄健康易感猪,2只试验猪在4 d内全部死亡;1986、1996、2006、2015年4个冻干代次的C43-5株菌种毒力和免疫原性检定结果均符合要求。同时提出了猪丹毒丝菌C43-5株毒力检定标准的修改建议,为延长菌种的保存期限提供了数据支撑。 In order to improve the virulence test standard of swine erysipelas vaccine,the growth rule of Erysipelothrix rhusiopathiae(E.rhusiopathiae)C43-5 strain in meat-liver-stomach digestion decoction was analyzed,the minimum lethal dose of the strain to pigs was determined,and the virulence and immunogenicity of 4 freeze-dried subcultures identified in 1986,1996,2006 and 2015 were tested.The results showed that the number of viable bacteria of C43-5 strain reached its maximum value when it was cultured for 16 hours in the decoction,then remained stable,but decreased rapidly after 19 hours,and a large number of bacteria died.Healthy susceptible pigs at 56~63 days old were intravenously injected with 3.5×10^(10) CFU live bacteria,2 experimented pigs died within 4 days;and the virulence and immunogenicity test results of C43-5 strain of the 4 freeze-dried subcultures were all qualified.Meanwhile,a recommendation on revision to the virulence test standard of E.rhusiopathiae C43-5 strain was put forward,supporting the extension of storage time of the strain.
