摘要
【目的】利用核酸适配体对氧氟沙星(OFL)的特异性识别功能及阳离子表面活性剂十六烷基三甲基溴化铵(CTAB)聚集金纳米粒子特性,建立一种简便、快速、灵敏的检测OFL的比色分析方法。【方法】利用柠檬酸钠还原法制备金纳米粒子,通过优化CTAB浓度和核酸适配体浓度,建立最佳试验条件,并对线性范围与特异性进行相关测定。【结果】经过优化得到反应的最佳试验条件为CTAB浓度0.95μmol/L,氧氟沙星核酸适配体浓度1.5nmol/L。在最优试验条件下,体系650 nm处与520 nm处吸光度比值ΔA650/A520与OFL浓度在5~75 ng/mL呈良好的线性关系,线性方程为:Y=0.0125X+0.0362(R^(2)=0.9883),最低检测限为0.295 ng/mL,该方法对OFL的检测具有较好的选择性。【结论】研究方法具有操作简便、快速、检测灵敏等优点,与其他方法相比具有更高的灵敏度,有望应用到实际检测当中。
[Objective]A simple,rapid and sensitive method for the colorimetric determination of Ofloxacin(OFL)was established by using the specific recognition ability of OFL by the aptamer and the aggregation of gold nanoparticles by the cationic surfactant cetyltrimethylammonium bromide(CTAB).[Method]Gold nanoparticles were synthesized by sodium citrate reduction.The assay conditions were optimized by optimizing CTAB concentration and aptamer concentration.The linear range and specificity were also determined.[Result]The optimal conditions were:CTAB concentration 0.95μmol/L and aptamer concentration 1.5 nmol/L.Under optimal conditions,the absorbance ratio ofΔA_(650)/A_(520) showed a good linear relationship with the concentration range of 5 ng/mL to 75 ng/mL of OFL,the linear equation was Y=0.012 5X+0.036 2(R^(2)=0.988 3).The limit of detection was 0.295 ng/mL and the method has good selectivity for the detection of OFL.[Conclusion]Compared with other methods,this method is simple,rapid and sensitive,and has a higher sensitivity.It is expected to be applied in practical testing.
