摘要
目的:应用多因素复合建模的方法构建血脂异常痰浊阻遏(PDR)证和脾肾阳虚(SKYD)证小鼠模型并验证该疾病-证候模型的质量。方法:以12只ApoE基因敲除小鼠构建疾病-证候动物模型(PDR组和SKYD组),以6只C57BL/6J小鼠作为正常对照组(NC组)。测定血脂指标,观察宏观行为,对血管内皮巨噬细胞进行转录组学分析。结果:在疾病模型方面,与NC组比较,PDR组和SKYD组TG、TC、LDL-C均显著增高(P<0.05);PDR组和SKYD组间TG、TC、HDL-C、LDL-C指标比较,差异无统计学意义;在证候模型方面,PDR组出现了痰浊阻遏的表现,SKYD组出现了脾肾阳虚的表现;在氧化应激反应方面,PDR组与SKYD组NF-κB信号通路和生物氧化反应差异均有统计学意义(P<0.01)。结论:本研究构建的病证动物模型符合相应病-证的特征性表现,具有较好的病证动物模型质量。
Objective:A multi-factor composite modeling approach was applied to construct mice models of dyslipidemia with phlegm-dampness retention syndrome and spleen and kidney yang deficiency syndrome and to validate the quality of the disease syndrome model.Methods:A total of 12 ApoE^(-/-)mice and establishment of disease-syndrome animal model by multi factor composite modeling method(PDR group and SKYD group).A total of 6 C57BL/6J mice were used as the normal control group(NC group).The indexes of blood lipid were measured,the macro behavior was observed,and the transcriptomic sequencing of vascular endothelial macrophages was analyzed.Results:The quality of disease animal models:Compared with the NC group,TG,TC and LDL-C in the PDR group and the SKYD group were increased(P<0.05).There was no significant statistical difference in TG,TC,HDL-C and LDL-C indexes between the PDR group and the SKYD group.The constructed quality of animal model of syndrome:From the macroscopic behavior,the mice in the PDR group showed the related manifestations of phlegm-dampness retention syndrome,and the mice in the SKYD group showed the related manifestations of spleen and kidney yang deficiency syndrome.The syndrome’s characteristics in animal model of oxidative stress:NF-κB signaling pathway and biological oxidation reaction were the different results which was closely related to oxidative stress(P<0.01).Conclusion:The disease-syndrome animal models constructed in this study conforms to the characteristic performance of corresponding disease and syndrome.The disease-syndrome animal models’quality is good.
