经皮穴位电刺激对卒中后抑郁大鼠行为学及前额叶皮质BDNF、CREB表达的影响 被引量:2

Effects of transcutaneous electrical acupoint stimulation on behavior and expression of BDNF and CREB in the prefrontal cortex of post-stroke depression rats

摘要 目的 观察经皮穴位电刺激(transcutaneous electrical acupoint stimulation, TEAS)对卒中后抑郁(post-strokedepression, PSD)大鼠神经功能、抑郁行为及前额叶皮质区脑源性神经营养因子(brain-derived neurotrophic factor, BDNF)、环磷腺苷效应元件结合蛋白(cAMP response element-binding protein, CREB)表达的影响。方法 48只Wistar大鼠随机分为空白(control, CON)组、脑缺血模型(middle cerebral artery occlusion, MCAO)组、卒中后抑郁模型(PSD)组、假针刺(sham acupuncture, SA)组、针刺(manual acupuncture, MA)组和TEAS组6组,每组8只。采用大脑中动脉闭塞与慢性不可预知的温和应激(chronic unpredictable mild stress, CUMS)法联合制备PSD模型,MCAO术后第3天于百会、内关和太冲进行穴位刺激。6组分别于干预第21天采用改良神经功能缺损量表(modified neuro-logical severity score, mNSS)评定大鼠神经功能损伤程度;蔗糖偏好实验、旷场实验评估大鼠抑郁行为学表现;PCR和Western blot法检测大鼠前额叶皮质区脑组织BDNF、CREB蛋白和mRNA表达。结果 21 d干预结束后,与CON组比较,其余各组大鼠m NSS评分均升高(P<0.01);与PSD、MCAO和SA组比较,MA组和TEAS组大鼠m NSS评分均降低(P<0.01)。与CON组比较,PSD组、MCAO组和SA组蔗糖偏好率均降低(P<0.01);与PSD组、MCAO组和SA组比较,MA组和TEAS组大鼠蔗糖偏好率均升高(P<0.01)。与CON组比较,PSD组、MCAO组和SA组移动距离和移动速度明显减少(P<0.01);与PSD组、MCAO组和SA组比较,MA组和TEAS组大鼠移动距离和移动速度明显增加(P<0.01)。与CON组比较,其余各组大鼠前额叶皮质BDNF与CREB mRNA和蛋白相对表达量均降低(P<0.01);与PSD组、MCAO组和SA组比较,MA组和TEAS组BDNF与CREB mRNA和蛋白相对表达量均升高(P<0.01)。结论 TEAS可明显改善PSD模型大鼠神经功能损伤和抑郁行为学表现,其机制可能与上调大脑前额叶皮质中BDNF与CREB mRNA和蛋白表达有关。 Objective To observe the effects of transcutaneous electrical acupoint stimulation(TEAS on neurological function, depressive behavior, and the expression of brain-derived neurotrophic factor(BDNF), cAMP response element binding protein(CREB) in prefrontal cortical areas of post-stroke depression(PSD) rats. Method Forty-eight Wistar rats were randomly divided into a control(CON) group, a middle cerebral artery occlusion(MCAO)group, a post-stroke depression model(PSD) group, a sham acupuncture(SA) group, a manual acupuncture(MA) group,and a TEAS group, with 6 rats in each group. The PSD model was prepared by middle cerebral artery occlusion combined with chronic unpredictable mild stress(CUMS), and acupoint stimulation was performed at Baihui( GV20),Neiguan( PC6) and Taichong( LR3) on the third day after MCAO. On the 21st day of intervention,the modified neuro-logical severity score(mNSS) was used to assess the degree of neurological impairment of rats in the six groups, respectively;Sucrose preference test, open field test was used to evaluate the behavioral performance of depression in rats;PCR and Western blot was used to detect the protein and mRNA expressions of BDNF and CREB in brain tissue from prefrontal cortex region of rats. Result At the end of the 21-d intervention, the mNSS score of THE rats in all other groups increased(P<0.01) compared with the CON group;the mNSS score of the rats in the MA group and TEAS group decreased(P<0.01) compared with the PSD group, MCAO group, and SA group. Compared with the CON group, the sucrose preference ratio in the PSD group, MCAO group, and SA group decreased(P<0.01);Compared with the PSD group, MCAO group, and SA group, the sucrose preference of rats in the MA group and TEAS group increased(P<0.01). Compared with the CON group, the moving distance and moving speed in the PSD group,MCAO group and SA group significantly decreased(P<0.01). Compared with the PSD group, MCAO group and SA group, the moving distance and speed of the rats in the MA group and TEAS group significantly increased(P<0.01).Compared with the CON group, the relative expression of BDNF mRNA and protein, CREB mRNA and protein in the prefrontal cortex of rats in all other groups decreased(P<0.01);Compared with the PSD group, MCAO group, and SA group, the relative expression amounts of BDNF and CREB m RNA and protein in the MA group and TEAS group increased(P<0.01). Conclusion TEAS can significantly improve neurological impairment and depressive behavior in PSD model rats, and the mechanism may be related to the up-regulation of BDNF and CREB mRNA and protein expression in the prefrontal cortex of the brain.
